Objective:To observe the effect of Xiao Xianxiong Decoction on the expression of ABCA1 and ABCG1 protein and cholesterol efflux in RAW264.7-derived foam cells,and to explore the mechanism of Xiao Xianxiong Decoction in preventing and treating atherosclerosisMethods:1.RAW264.7 macrophages were treated with different concentrations of ox-LDL to prepare foam cell model.Oil red O staining and qRT-PCR were used to identify and screen the appropriate concentration of ox-LDL2.RAW264.7-derived foam cells were plated in 96-well plates and incubated with different concentrations of Xiao Xianxiong Decoction for 24 hours.The cell survival rate was detected by CCK-8 kit,and the appropriate concentration of Xiao Xianxiong Decoction was selected to interfere with the cells.3.RAW264.7-derived foam cells were intervened at high,middle and low concentrations of Xiao Xianxiong Decoction for 24 hours,and the lipid accumulation and morphological changes of foam cells were detected by oil red O staining.4.The expression of ABCA1 and ABCG1 proteins in each group was determined by western blot5.NBD-cholesterol combined with fluorescence enzyme assay was used to detect the effect of Xiao Xianxiong Decoction on intracellular cholesterol effluxResults:1.The foam morphology of the cells treated with 25 ?g/ml ox-LDL was typical and the damage was moderate,which was selected as the modeling concentration for the follow-up experiment2.With the increase of the concentration of Xiao Xianxiong Decoction,the cells survival rate showed a downward trend,and reasonable concentrations of 2.5 mg/ml,1.25 mg/ml and 0.625 mg/ml were selected as high,medium and low concentrations in the later experiment3.Compared with the model group,Xiao Xianxiong Decoction in high and middle concentration groups could significantly reduce the intracellular lipid accumulation4.Compared with the model group,Xiao Xianxiong Decoction in high and middle concentration groups can significantly increase the expression of ABCA1 and ABCG1 protein in foam cells5.Xiao Xianxiong Decoction in 1.25 mg/ml concentration group can significantly promote cholesterol effluxConclusion:Xiao Xianxiong Decoction can reduce lipid accumulation and promote cholesterol efflux by up-regulating the expression of ABCA1 and ABCG1 protein in RAW264.7-derived foam cells,so as to play the effect of clearing phlegm,thus slowing down the occurrence and development of AS.
