| Objective: Many epidemiological and experimental studies found that lycopenecould reduce the risk of many types of cancer danger.The research project topic oflycopene on cell proliferation and apoptosis effects of lycopene on human gliomaU251cells in vivo and to explore the mechanism of action.Lycopene as a naturalchemical prevention agent use for dietary prevention,to reduce or delay preventglioma further used in clinical treatment provide theoretical foundation.Methods:The U251cells were sintered DMEM nutrient solution (including100u/ml penicillin,0.2%NaHCO3and10%fetal calf serum),in5%CO2,37℃incubator foster.1.Observe the effect of different concentrations of lycopene forms atdifferent times of the descendants of glioma cells.2.Use CCK-8method determinesurvival rate.3.Use trypan blue staining measure growing curve of the differentgroups of people with glioma cell.4.By flow cytometry detection cell cycle andproliferation index(proliferation index,PI)=(S+G2M)/(Go/Gl+S+G2M)×100%iscalculated.5.By SCAR marker dye tracer technique understand the effects of lycopeneon cell intercellular communication.Result:1.After treatment of cell stable by different concentrations of lycopene,then using CCK-8method to detect cell survival rate every day, the results showedthat lycopene have certain effect on human glioma cell proliferation that has therelationship with the dose.2.Human brain glioma cells treated by lycopene havevarying degrees of inhibition the more dose of lycopene, the more inhibiting effect.3.G0+G1phase ratio increases, S phase and G2+M phase (SPF isS Phase Fraction)ratio decreases by different concentrations of lycopene。In the roleof time under the same conditions, proliferation index (PI) and S phase proportionwith the increase of concentration of all groups cells culture were decreased.4.Withthe increase of concentration of lycopene, LY transmission distance increases. there isan obvious dose-effect relationship between them.Summary:Experiments of Lycopene on the U251cells of the human brain vitroglioma confirmed that as follows:1.U251cells can be induced to apoptosis andregulate the cell cycle, tumor cells in Gl/S by Lycopene.2.Intercellular spaceconnection communication function between glioma cells can restoration. But thecorrelation study of lycopene antitumor is less now.Further study will explore thelycopenes’ other influence mechanism of the human brain glioma and as the naturalchemical prevention agent in dietary intervention to reduce or delay the glioma,thathas important practical significance if it can be applied to clinical therapy. |
