Objectives:To studied the changes in the characteristics of incipient peripheral blood T cell subsets (CD4+, CD8+, CD4+/CD8+) in children with TIDM.To explored the correlation between the levels of incipient peripheral blood T cell subsets and pancreatic islet function.Methods:43patients with incipient TIDM in Shanxi Children’s Hospital from2010.5-2013.12were enrolled and25healthy subjects were selected as control group.FCM was used to detecting serum levels of T cell subsets(CD4+,CD8+,CD4+/CD8+).Random blood glucose、FPG、C-P、 insulin and HbAlc also were detected and compared among the two groups.The correlation analysis was carried on the changes of T cell subgroups and the levels of HbAlc and C-P in patients with incipient TIDM.Results:1.Compared with control group,the levels of random blood glucose(21.49±7.85mmol/L) FPG(12.07±4.52mmol/L)、HbAlc(12.85±2.38%) were significantly higher in group TlDM(f=13.41,f=10.12,t=11.95;pall<0.05).However,the levels of Ins(1.98±1.42mIU/L)、C-P(2.29±0.19ng/ml) was significantly lower in group TIDM (t=12.58,1=14.82, pall<0.05).2.Compared with control group,the levels of CD4+T cell subsets (36.39±8.47%)、 CD4+/CD8+T cell subsets(1.46±0.66%) were significantly higher in group T1DM(1=13.41,t=10.12,t=11.95;pall<0.05).CD8+Tcell subsets(26.15±6.70%)significantlylower(f=1.531,p<0.05)3.In group TlDM,the change of CD4+T cell subsets was positively related to the level of HbAlc(r=0.261, p<0.05),but negatively correlated with C-P(r=-0.270, p<0.05);the change of CD8+T cell subsets was no obvious correlation with the level of HbAlc((r=-0.156,p>0.05)and positively correlated with C-P(r=0.162, p<0.05).There was no obvious correlation between CD4+/CD8+T cell subsets and the level of HbAlc(r=0.200, p>0.05),but was negatively correlated with C-P(r=-0.243, p<0.05).Conclusions:1. In balance of T cell subsets is involved in onset procedure of TlDM,mainly for abnormal activation of CD4+T cell subsets,CD8+T cell subsets defect and CD4+/CD8+ratio increased.2. The change of cellular immune function is closely related to the damaged islet beta cells.CD4+T cell subsets and CD4+/CD8+T cell subsets are negatively related to insulin function, CD8+are positively correlated with insulin function.The change of cellular immune function will point out the damage of slet beta cells,as well as the levels of CD4+T cell subsets rising and CD8+T cell subsets reducing.
