Objective Discuss the p38MAPK inhibitor in myocardial injury and short-termeffects to SD rats,by using CBS3830pretreatment of rats before cardiopulmonarybypass(CPB).Methods Selecting32male SD rats (body weight350-500g). Randomly assignedcontrol group(group A,n=8), Pseudo surgery group(group sham,n=8), cardiopulmonarybypass(groun CPB,n=8),IDO Inhibitors of preprocessing grou(pgroup B,n=8). GroupB IDO Inhibitors of preprocessing group preoperative oral gavage IDO inhibitors1-methyl-d-tryptophan100mg/(KD.d), seven days in a row. Group A,Group sham andgroup CPB given the same volume of saline solution in preoperative oral gavage forseven consecutive days. Seven days later, group CPB and group B use10%chloral0.3ml/kg by intraperitoneal injection of anesthesia. Though right jugular vein intubationdrainage. Tail artery infusion. The left femoral artery catheterization monitoring bloodpressure,build minimally invasive cardiopulmonary bypass model in rats. Filling flow80-00ml/(kg.min). cardiopulmonary bypass time is60min. Group sham adopted thesame way anesthesia, only for intubation and heparin. Group A narcotic in the samemanner, without intubation, directly sampling after the heparinized for later use.Preoperative, intraoperative, and postoperative monitoring of blood pressure, blood gas.Maintenance of rats ’ vital signs are stable. All rats were successfully concluded duringcardiopulmonary bypass. Postoperative survival in rats maintained24h. Respectivelysampling blood4ml, after the catheterization, after the end of bypass, aftercardiopulmonary bypass24h. After centrifuge15min5000rpm take for upper plasmamonitoring IDO,CRP,TNF-α and IL-6concentration. Results1.All rats were successfully completed surgery, surviving24h, blood pressure,heart rate and arterial blood gas remain stable.2. Preoperative,postoperative,post-operative24h take in rat plasma at-80℃after saving targets, display aftercardiopulmonary bypass time IDO concentration and inflammatory markers in plasmaincreased significantly.3. The IDO concentration of IDO inhibitors pretreatmentgroup is decreased compare with CPB group, the in flammation indicators will also bereduced.Conclusion1.The right jugular vein cannulation drainage, tail artery infusion, the left femoral artery catheter monitoring blood pressure successful build minimally invasive cardiopulmonary bypass models in rats.2. Cardiopulmonary bypass causing systemic inflammatory response.3. Cardiopulmonary bypass can cause IDOincreased,。IDO as monitoring an indicator of inflammatory response during cardiopulmonary bypass.4. Specificity of inhibition activity of IDO in rats by lessening the inflammation responses in cardiopulmonary bypass.
