Background and ObjectiveThe treatment of MDD at present mainly adopts5-HT reuptakeinhibitors (SSRI). In the affirmation of that new type of antidepressantresearch, and at the same time, its curative effect, side effects and theway of delivering still need improving. Vanillin molecule has a creamyflavor. Research shows vanillin has the effect of anti-anxiety,anti-bacteria and anti-oxidation. What are the protein targets whichexist in the nature or in the vanillin of human body?Olfactory pathway has a wide range of fiber links to the emotioncenter amygdala and hippocampal neuron which makes it becomes thetheoretical basis of the olfactory pathway regulating emotional center.More and more reports are involved in the treatments of CNS byolfactory pathway. The administration route of olfactory pathway isconvenient and has no peripheral side effects. The smell of vanillin hasthe effect of anti-anxiety. Does vanillin molecule’s "pleasant" odor havean intervention effect on the patients of MDD? If it does, what are theunderlying mechanisms? This researcher is to explore theanti-depressive effects of vanillin by sniffing and underlyingmechanism. The reverse virtual screening of vanillin molecular is toexplore the possible role of involved receptors in MDD and medicinalvalue of vanillin. Part One The Reverse Virtual Screening of VanillinMedicinal TargetsMethodsThe combination of drugs and protein is similar to theidentification of a key and lock. In order to research the receptor thatvanillin could work on and its’ potential value, we use moleculardocking software Surflex to screen sc-PDB protein database. Thesoftware score automatically to the results, the higher the score ofresults, the correlation between protein and vanillin molecular is higher.This virtual screening is valuable to the study of the vanillin receptorsand has guiding significance in development of medicinal.ResultMore than eight points of the protein crystal are:Carboxyethyl-arginine beta-lactam-synthase, Metallo Beta-lactamase,M. tuberculosis Salicylate Synthase, Beta-lactamase IMP-1. This isconsistent with reports of vanillin antibacterial and antioxidant action,and the top20protein crystals are shown in table1-1.Part Two The Influence of Vanillin Sniffing onBehavior of MDD RatsMethodsCUMS+solitary and OBX ways were used to support theestablishment of depression model. All rats were divided into5groups,10in each. They were CUMS+Vanillin; CUMS+Fluoxetine; Model;OBX+Vanillin and Sham group. The behavior were observed pre andpost-CUMS (or operation) ResultsIn CUMS+Vanillin and CUMS+Fluoxetine group, after4weeks’treatment, the behavior improved significantly, while OBX+Vanillinand Model group were no significant improvements.Part Three The Influence of Vanillin Sniffing on5-HT; DA and NE of MDD RatMethodsThe behavior observations have shown that vanillin can improvedepressive behavior in rats via the olfactory pathway. In order toexplore its mechanism, quantitative determination of monoamineneurotransmitters in brain were carried out by ultrahigh performanceliquid chromatographic technique in order to find the underlyingpossible mechanisms. The total soluble protein in supernatants wasconsidered as the internal reference standard of monoamineneurotransmitters. It was detected by BCA protein assay kits.ResultsIn CUMS+Vanillin group, after4weeks’ treatments, the level of5-HT; DA were significantly increased compared with Model group, NEwere no significant changes; In OBX+Vanillin group, the level of5-HT;DA; NE were all decreased significantly compared with Sham group.Conclusion1The result of reverse virtual screening of vanillin medicinal targetsis consistent with reports of vanillin anti-bacteria and anti-oxidation.2Vanillin may relief the depression-like behavior in rats by olfactorypathway;3The mechanism may be increasing the level of brain5-HT and DA;...
