Objective To detect differentially expressed proteins in rat brain cortextissue by two-dimensional gel electrophoresis (2-DE) and mass spectrometry(MALDI-TOF-MS) techniques, thus to explore and analyze the differenceprotein expression in the cerebral cortex that gabapentin (GBP) acts onvincristine(VCR)-induced neuropathic pain.Methods36SD rats were randomly divided into3groups:①Blankcontrol group (Group Ns): Odd day, intraperitoneal injection of NS; even day,gavage NS;②Pain model control group (Group Pc):Odd day, intraperitonealinjection of VCR; even day, gavage NS;③Gabapentin treatment group (GroupGt): Odd day, intraperitoneal injection of VCR; even day, gavage GBP.Administration is total16days. During the administration, the rat behaviorchange, body weight and heat pain threshold were observed. Afteradministration, sciatic nerve ultrastructural changes shall be observed with lightmicroscope and electron. The differentially expressed proteins in cortex tissue ofrat were separated by2-DE, then identified by MALDI-TOF-MS.Result1. Body weight: At the12th days and16th days, the rats weight ofgroup Pc compared with group Ns and group Gt in the same period decreased,the difference is statistically significant(p<0.05); comparing the16th days and before the experiment in three groups, weight decreased, the difference isstatistically significant (p<0.05).2. Heat pain threshold: Heat pain threshold ofgroup Pc after adminstration dereased comparing with group Ns and group Gt inthe same period and group Pc before adminstration, the difference is statisticallysignificant (p<0.05).3. sciatic nerve tissue ultrastructure: Group Ns was normalrat sciatic nerve fiber structure; Group Pc was significant degeneration; GroupGt was slighter degeneration than Group Pc, even close Group Ns.4. Twodimensional gel electrophoresis and mass spectrometry: Compared with GroupPc, there are15difference proteins in Group Gt, then identified9kinds ofproteins: DJ-1, ATP synthase subunit D, phosphatidylethanolamine bindingprotein-1(PEBP1), Hemoglobin subunit alpha-1/2, Majorbeta-hemoglobin,Cu-Zn superoxide dismutase (SOD1), peroxidase-2(Prx-2), nucleosidediphosphate kinase A(NDKA), heat shock protein10(HSP10).Conclusion Gabapentin affect protein expression of vincristine-inducedneuropathic pain in rat cerebral cortex, there may be regulatory mechanism ofcentral protein levels.
