Objective: To investigate the effect of compound Shichangpu (CSCP) onepilepsy model induced by pentylenetetrazol (PTZ).Methods:(1) The epileptic seizure of mice induced by PTZ was used toevaluated the protective effect of CSCP on behavior, structure changes ofhippocampus. Sixty Kunming mice, including half male and half female, wererandomly divided into6groups as follow: a normal control group, model group,phenytoin (PHT) group, high-dose CSCP (4000mg·kg-1) group, middle-doseCSCP (2000mg·kg-1) group and low-dose CSCP (1000mg·kg-1)groups. Thenormal control group and model group were given intragastric administrationsame volume of saline, expect the normal control group, the mice in the groupswere injected intraperitoneally with PTZ (80mg·kg-1) to induce the acuteepilepsy model. The influence of CSCP on incubation period,behavior degree,times of epileptic seizure among PTZ induced epilepsy mice, the structurechanges of mice were detected by the brain tissue HE staining.(2)The effect ofCSCP on chronic kindling rat model was also analyzed:①the CSCP on thebehavior in epileptic rat was deteced the behavior, incubation period and the times with epileptic;②The activities of SOD, T-AOC and the contents of MDAin brain cortex tissue were analyzed, respectively;③The mRNA levels ofMDR1, MRP1and Caspase-3in hippocampus tissue were determined byFQ-PCR assay;④Detecting CSCP on GFAP, GABA and Glu of chronic kindlingrat hippocampus expression by immunohistochemistry(IHC)technology.Results:(1) the effect of CSCP on behavior, structure changes of hippocampusin epileptic seizure of mice was deteced the behavior: the results showed thatCSCP can improve the degree of seizure attack, prolong the incubation periodof mild attack, and decrease Ⅳ and Ⅴ degree epileptic seizure, the PTZinduced epilepsy mice could damage the structure of hippocampus, reduce thenumber of cells, PHT and CSCP could ameliorate these changes.(2) The effectof CSCP on epileptic rat induced by pentylenetetrazol(PTZ):①The ratchronic epilepsy model was by intraperitoneal injection of pentetrazole(PTZ), itswere successfully kindling. Higher dose of CSCP could improve the degree andtimes of epileptic seizure remarkably (P﹤0.05or P﹤0.01);②Compared withthe model control group, the activities of SOD and T-AOC in cytopiasm of therat brain cortex tissues decreased while MDA increased.(P﹤0.05or P﹤0.01);③Compared with the model control group, the mRNA expressions of MDR1,MRP1and Caspase-3were down-regulated in the CSCP groups.(P﹤0.05or P﹤0.01);④Compared with the model control group, GFAP overexpression canbe inhibited by CSCP, its can elevated GABA of hippocampus of rats, andreduced Glu expression.(P﹤0.05or P﹤0.01).Conclusions:CSCP could inhibit the epileptic seizure in mice induced by PTZ. In chronickindling rats induced by PTZ, CSCP has certain treatment function, which the mechanisms may be related to attenuating free radical, inhibiting the lipidperoxidation, decreased the multiple drug resistance gene expression andmodulated the balance of excitatory and inhibitory neurotransmitter.
