PTEN Expression And Its Clinicopathological Significance In Gastric Cancer:a Meta-Analysis

Objective To investigate the relationship with clinicopathologic features between PTEN (phosphatase and tensin homolog deleted on chromosome ten, also called MMAC1, mutated in multiple advanced cancers1) expression and gastric cancer.Methods The published studies were searched in Cochrane Library, PubMed, EMbase, CNKI, VIP, WanFang Database and CBM from inception to July24th,2013,and other relevant references of the included literature were also searched to collect all the case-control studies. Two reviewers independently screened studies according to the inclusion and exclusion criteria, extracted data and assessed quality of the included studies independently.Then meta-analysis was conducted using RevMan5.2software.Results A total of14studies were recruited,which included1026cases in the gastric cancer group and539cases in the normal gastric mucosa group. The results of meta-analysis showed that: PTEN expression was lower in the gastric cancer group than the normal gastric mucosa group, with a significant difference (OR=0.07,95%CI=0.05-0.10,P<0.00001). No significant difference was tested between age (OR=1.34,95%CI=0.88-2.04, P=0.17),and sex (OR=0.91,95%CI=0.60~1.36, P=0.64).There were significant differences between tumor length (OR=3.67,95%CI=2.02~6.67, P<0.0001), histologic subtype (OR=3.54,95%CI=2.56-4.91P<0.00001) invasion depth (OR=3.31,95%CI=2.37-4.63,P<0.00001),lymphatic metastasis (OR=4.11,95%CI=2.98~5.67, P<0.00001),and TNM stage (OR=3.94,95%CI=2.74~5.65, P<0.00001) respectively.Conclusion PTEN expression in gastric cancer group was lower than that in normal gastric mucosa group. It has nothing to do the age and sex of patients and with the relevant of tumor length, histologic subtype, invasion depth, lymphatic metastasis and TNM stage. Because the small sample size and included trials only contained English and Chinese, more large sample and high-quality studys are required to define the relationship of PTEN and gastric cancer.
