OBJECTIVETo investigate the effect of exogenous hydrogen sulfide on intestinal fibrosis induced by trinitrobenzene-sulfonic acid?TNBS?in rats.METHODSSixty healthy femal Sprague-Dawley rats were randomly divided into 4groups:alcohol-control group,model group,NaHS low-dose and high-dose group.Intestinal fibrosis were induced by TNBS in model group,NaHS low-dose and high-dose group.At the beginning of the third week,low-dose and high-dose NaHS groups were injected intraperitoneally once a day with NaHS?14.4umol/kg and 36.0umol/kg,respectively?,an equal volume of normal saline was injected in alcohol-control and model groups.General situation was observed daily during the experiment.All rats were killed at the end of the fifth week.Serum H2S level was measured.Their colons were resected for the macroscopic and microscopic scores and collagen area,CollagenI?2?Collagen??TGF-?1?TNF-?were detected by Elisa method,CBS mRNA expression was determined by RT-PCR.RESULTS1)The macroscopic score in model group and NaHS groups was higher thanalcohol-control group.The macroscopic score in NaHS groups was lowerthan model group,while the parameter in high-dose NaHS group wassignificantly lower than low-dose NaHS group?P<0.05?.2)The microscopic score and collagen area in model group and NaHS groupswere higher than alcohol-control group.The microscopic score and collagenarea in NaHS groups were lower than model group,while the parameters inhigh-dose NaHS group were significantly lower than low-dose NaHSgroup?P<0.05?.3)The level of Collagen I?2?Collagen III?TGF-?1?TNF-?in model group andNaHS groups were higher than alcohol-control group.The level ofCollagen I?2?Collagen III?TGF-?1?TNF-?in NaHS groups were lowerthan model group,while the parameters in high-dose NaHS group weresignificantly lower than low-dose NaHS group?P<0.05?.4)The level of CBS mRNA in model group and NaHS groups was lower thanalcohol-control group.The level of CBS mRNA in NaHS groups washigher than model group,while the parameter in high-dose NaHS groupwas significantly higher than low-dose NaHS group?P<0.05?.5)The level of serum H2S in model group and NaHS groups was lower thanalcohol-control group.The level of serum H2S in NaHS groups was higherthan model group,while the parameter in high-dose NaHS group wassignificantly higher than low-dose NaHS group?P<0.05?.CONCLUSIONSExogenous hydrogen sulfide was closely related with TNBS induced intestinal fibrosis in rats,and has anti-inflammatory and anti-fibrotic effects within in a dosage—dependent manner.
