Abstract: objective: this study was designed by detecting p-STAT3, p-AKT, p-mTOR,p-ERK1/2expression in Non-small cell lung carcinoma, and to study the pathogenesis andrelationship of p-STAT3, p-AKT, p-mTOR, p-ERK1/2between the occurrence anddevelopment in NSCLC, in order to further clarify the p-STAT3,p-AKT,p-mTOR,p-ERK1/2protein expression and regulation,and provide a theoretical basis for prevention andtreatment of NSCLC.Materials and Method:49cases of NSCLC surgery specimens and19casesnon-cancerous tissues surrouding tumors were collected in First Hospital of Jilin Universityfrom january to august2013.None of these patients was treated by chemotherapy orradiotherapy before surgery. their clinical information of these patients include age, sex,pathological type, smoking status, TNM stage, and lymph node metastasis was got bysearching the medical records. SP immunohistochemical staining method of p-STAT3, p-AKT,p-mTOR, p-ERK1/2expression in NSCLC and non-cancerous tissues surrouding tumors.statistical analysis was performed using SPSS statistical software package(19.0).P values≤0.05indicated statistically significant difference.Results1.The positive rate of p-STAT in NSCLC was71.4%(35/49),the difference ofexpression of p-STAT3between the NSCLC and nomal lung tissue shows statisticallysignificant(p<0.001).The express positive rate of p-STAT3was different in pathologicalcharacteristics whereas no diffenrent were seen considering age, sex, smoking, TNM stage,total stage.The positive rate of p-STAT3was different in adenocarcinoma, squamous cellcarcinoma (45.0%vs89.7%, p=0.001).2. For49NSCLC patient speciments,the positive rate of p-AKT was75.5(37/49),thedifference of expression of p-AKT between the NSCLC and nomal lung tissue showsstatistically significant (p<0.001).The express positive rate of p-AKT has no different were seen considering sex, age, smoking, pathological, TNM stage, total stage.3. The positive rate of p-ERK1/2in NSCLC was83.6%(41/49),the difference ofexpression of p-ERK1/2between the NSCLC and nomal lung tissue shows statisticallysignificant(p<0.001).The express positive rate of p-ERK1/2was no different in sex, age,smoking,pathological,TNM stage,total stage.4. The positive rate of p-mTOR in NSCLC was75.5%(37/49),the difference ofexpression of p-mTOR between the NSCLC and nomal lung tissue shows statisticallysignificant(p<0.001).The express positive rate of p-mTOR was different in N stage whereasno diffenrent were seen considering age, sex, smoking, T stage, M stage, total stage,pathological characteristics. the positive rate of p-mTOR was different in N stage (p=0.043).5. In NSCLC tissue p-STAT3and p-ERK1/2protein expression was positivelycorrelated (p=0.033), and p-ERK1/2and p-mTOR protein expression was positivelycorrelated.Conclusions1. p-STAT3, p-AKT, p-ERK1/2, p-mTOR are high express in NSCLC.2. The express rate of p-STAT3in squamous cell carcinoma was higher than inadenocacinoma.3. the positive rate of p-mTOR was higher in lymph node metastasis speciments.4. p-STAT3and p-ERK1/2, p-ERK1/2and p-mTOR were positively correlated with eachother.
