Objective:Explore the mechanism of oligodeoxynucleotide MT01(Anoligodeoxynucleotide designed based on human mitochondrial DNAsequence) into the human periodontal ligament cells, and provide anexperimental evidence for oligodeoxynucleotide MT01as a drug using oninhibition of alveolar bone absorption.Methods:Cultured and identified human periodontal ligament cells in vitro. Humanperiodontal ligament cells and Cy5-labeled oligodeoxynucleotide MT01(working concentration1.0μg/ml)were incubated for15minutes,30minutes,1hour,3hours,6hours,12hours,24hours,36hours and48hours. Laserconfocal scanning microscopy traced and recorded the situation of MT01entering human periodontal ligament cells at nine time points, and comparativeanalysised MT01fluorescence intensity in human periodontal ligament cells atdifferent time points. Using statistical software of SPSS17.0and pared-samples t-test, analysis experimental datas between two groups. P <0.05wasstatistically significant, and P <0.01was statistically significant difference.Results:Fluorescence intensity of1.0μg/ml MT01into human periodontalligament cells at15minutes,30minutes,1hour,3hours,6hours,12hours,24hours,36hours and48hours, respectively:1484±128,1771±206,2773±1364,1874±121,2437±293,3170±297,2442±273,2285±45,2119±374.12hours in the experimental group in which the fluorescence intensity for15minutes,30minutes,6hours,48hours in the experimental group compared tothe fluorescence intensity were significantly different (P <0.01). There were nosignificant differences among the other groups. Conclusion:1. Oligonucleotide MT01enter the cytoplasm of human periodontalligament cells through rapid endocytosis without the need for vectorsand certain concentration of oligonucleotide MT01enter humanperiodontal ligament cells was time-dependent:12hours reach thepeak.2. Oligonucleotide MT01distribute at bipolar of human periodontalligament cells.
