Background/Aims:The increasing number of nonacoholic fatty liverdisease has taken place in recent years,its pathogenesis is too complicated,and miR-122has a high degree of specificity and expression in liver. Somestudies suggest miR-122is related to viral hepatitis type C,nonacoholic fattyliver disease,hepatocelluar carcinoma and so on,but there are somearguments in the study of miR-122’ expression in NAFLD,we infer it maypromote the pathogenesis of NAFLD,and analyze the expression of miR-122in NAFLD.Methods: According to the diagnostic and treatment guidelines ofNAFLD of China,2010,we enrolled a total of41cases of NAFLD patient inSichuan Provincial People’s Hospital from March2012to October2012.Thepatients all knew the fact and agreed,and their livers had biopsy-provenNAFLD.Patients were divided into two groups:10cases in NAFL group and31cases in NASH group.And20healthy controls matched for age,genderwere recruited in the normal control group.We collected their generalinformation,biochemistry and metabolism index,extracted total RNA of liverusing Trizol Reagent according to the manufacturer’s instructions andvalidated miR-122difference expression in liver by quantitative RT-PCR,therelative expression was calculated using the comparative ΔΔCTmethod,andfinally statistically analyzed measurement consequence in NAFLD group andnormal group,as well as in NAFL group and NASH group by SPSS15.0 software.Results:①Compared to normal group, there was a statisticalsignificance of body weight,waist circumference,hip circumference,BMI,waist/hip ratio, blood pressure in NAFLD group(P<0.05),especially in bodyweight,waist circumference,BMI, waist/hip ratio and systolic pressure((P<0.001).②There was a statistical significance of serum ALT,AST,GGT,UA,TC,TG,LDL-C,FBG,FINS,HOMA-IR and IAI in NAFLDgroup comparing to normal control group respectively (all P<0.05),whilethe differences of ALT,TC,LDL-C,FBG,FINS,HOMA-IR and IAI hadnotable statistical significance(p<0.001).However,there was no statisticalsignificance of Alb, TBIL, HDL-C between NAFLD and normalgroup(P>0.05).③Compareing NAFL group to NASH group,there was nostatistical significance in above mentioned measurement consequence(P>0.05).④There was notable statistical significance of the miR-122’srelative expression in NAFLD group comparing to normal group (P<0.001),but the difference had no statistical significance in NAFL group and NASHgroup (P=0.225>0.05).⑤We analyzed diagnostic value of miR-122inNAFLD using ROC area under curve,the AUC value is0.963,its sensitivityand specificity is85.4%and95.0%respectively.Conclusion:The present study demonstrates that miR-122is stronglyupregulated in non-alcoholic fatty liver disease,suggests an important role ofmiR-122in hepatic lipid metabolism and implicates the participation ofmiR-122in the pathophysiological processes of non-alcoholic fatty liverdisease.These results broaden our biological understandings of NAFLD and offer a new direction for future clinical of miR-122as applications potentialtargets....
