Corydalis Ochotensis Turcz is the perennial herb of Corydalis genus Papaver; thealiases: Huanglong shelling, gas grass; it is mainly distributed in Jilin, Heilongjiang, HebeiProvince and other places in China. This herb can be wholly used on dysentery, sorhurts,hemoptysis, phthisis pulmonary boils and so on. At present, domestic and foreign scholarshave more research on chemical composition of the genus Corydalis, butlittle research onCorydalis Ochotensis Turcz. Therefore, for further researchand development of CorydalisOchotensis Turcz, this paper studies on it as following aspects:1. In this article alkaloids of Corydalis Ochotensis Turcz were summarized; andalkaloids on the anti-tumor and anti-Alzheimer’s were reviewed.2. Alkaloid chemical components of Corydalis Ochotensis Turcz were extracted andseparated. the whole plant after crushing was percolation extracted with0.5%sulfuric acidsolution, and purificated with AB-8macroporous resin. The total alkaloids after purified wasextracted by silica gel column chromatography, preparative thin layer silica gel column andrecrystallization. As a result, six compounds were obtained. According to theirphysicochemical properties, MS and NMR, combined with the literatures, we identified thecompounds were Protopine, ochotensimine, fumariline, sanguinarine, tetrahydroberberineand berberine.3. The content of protopine in Corydalis Ochotensis Turcz and total alkaloids wasdetermined by HPLC, using C18column, acetonitrile-triethylamine acetate solution,(taketriethylamine8ml, dilute acetic acid30ml, add water to1000ml)(18:82) as the mobile phase;detection wavelength of289nm. The method is simple, with good precision andreproducibility and reliable for quality control of Corydalis Ochotensis Turcz.4. In order to evaluate the therapeutic effect of the total alkaloid to AD rats which wereinduced by β-amyloid protein, the Morris water maze test was adopted. Experimental resultsshow that low-dose, medium-dose and high-dose of the total alkaloids can improve learningand memory function of β-amyloid dementia rats when the hydrochloride donepezil aspositive control.5. The vitro anti-tumor activity of the three monomers alkaloid compounds: protopine,Fumriline, sanguinarine were researched by MTT. The results showed Protopine and sanguinarine on have good antitumor activity on Hep2(larynx epidermoid carcinoma),HepG2(liver cancer cells), SW480(colon cancer cells) and A549(lung cancer cells).In summary, We study on Corydalis Ochotensis Turcz chemical constituents and sixcompounds (protopine、ochotensimine、fumarilin、sanguinarine、etrahydroberberine andberberine) were isolated and identified. In Corydalis Ochotensis Turcz quality research, thedetermination of protopine in Corydalis Ochotensis Turcz was established by HPLC. In theactivity study of the alkaloids in Corydalis Ochotensis Turcz, the therapeutic effect of thealkaloids to AD rats which were induced by β-amyloid protein was evaluated by the Morriswater maze was investigated. And the vitro antitumor activity ofthe three monomers alkaloidcompounds were researched by MTT. The topic provide a scientific basis for further researchon Corydalis Ochotensis Turcz.
