Objective:The goal of this article is to research MACC1and OCT4gene detected byimmunohistochemistry, in normal endometrium, simple and complex hyperplasia, atypicalendometrial hyperplasia and endometrial adenocarcinoma, and research the expression levelin different tissues above situation with histopathological grading, surgical-pathologic stage,and lymph node metastasis by statistical test method. Explore their role in the occurrence,development and prognosis of endometrial adenocarcinoma, and to provide a reference forthe early diagnosis of endometrial adenocarcinoma, treatment and condition assessment andprognostic evaluation.Methods:Selected the tissue specimens by surgical resection in The China-Japan Union Hopsitalof Jilin University and The Central Hospital of Changchun between September2011andSeptember2013. Select the specimen were freshly drawn immediately fixed in10%neutralformalin solution, paraffin embedded. Slice4μm thick, stained by HE,MACC1and OCT4immunohistochemical staining. Positive control for known positive sections contained in thekit, pathologists identified all slices by blinded, immunohistochemistry staining tissue todetection MACC1and OCT4expression.Results:1. MACC1protein positive particles brownish yellow, located in the cell membrane and(or) cytoplasm. The positive expression of MACC1in normal endometrium, simple andcomplex hyperplasia, atypical endometrial hyperplasia and endometrial adenocarcinoma is20%、33.33%、40.0%、53.13%,and75%.Compared among the five groups, P<0.01,therewas a significant statistical difference. MACC1protein expression was significantly elevated in endometrial adenocarcinoma, there was a statistically significant difference comparedwith the remaining four groups, there were statistically significant of every compare.Compared among the first four group, P>0.05, there was no significant difference orstatistically significant.2. OCT4protein positive particles brownish yellow, located in the nucleus. The positiveexpression of OCT4in normal endometrium, simple and complex hyperplasia, atypicalendometrial hyperplasia and endometrial adenocarcinoma is26.67%、36.67%、40.00%、56.25%and77.08%. Compared among the five groups, P<0.01,there was a significantstatistical difference. OCT4protein expression was significantly higher in endometrialadenocarcinoma, there was a statistically significant difference compared with the remainingfour groups. Compared among the first four group, P>0.05, there was no significantdifference or statistically significant.3. The positive expression rate of MACC1protein is higher in surgical stage III~IVthan I~II. Compared between the two group, P<0.05, there was a statistically significant.Compared between the two group, in histopathological grading G1+G2and G3, P<0.05,there was statistically significant. The positive expression rate of MACC1protein in withoutlymph node metastasis was72.73%, and lymph node metastasis was37.93%. Comparedbetween the two group, P<0.05, there was statistically significant.4. The positive expression rate of OCT4protein is higher in surgical stage III~IVthan I~II,respectively76.92%,40.0%. Compared between the two group, P<0.05, therewas a statistically significant. Compared between the two group, in histopathologicalgrading G1+G2and G3, P<0.05, there was statistically significant. The positive expressionrate of OCT4protein in without lymph node metastasis was44.83%,and lymph nodemetastasis was81.82%,Compared between the two group, P<0.05, there was statisticallysignificant.5. The positive expression rate of MACC1and OCT4in endometrial adenocarcinomawas:75%、77.08%,it is postive correlation between MACC1and OCT4positive expressionin endometrial carcinoma(r=0.258,P=0.039,P<0.05).Conclusion:1. MACC1and OCT4in the positive expression rate in malignant change from normaltissue to the whole process is gradually increasing, suggest high expression of MACC1and OCT4in endometrial adenocarcinoma play an important role in occurrence and development,can be used as a judgement of diagnosis endometrial adenocarcinoma.2. MACC1and OCT4high expression in endometrial adenocarcinoma is associatedwith Operation pathologic staging,histological grade and lymphatic metastasis or not,theprompt can be used as prognosis of endometrial adenocarcinoma.3. MACC1and OCT4in endometrial adenocarcinoma in the expression showed apositive correlation, the joint detection can improve the diagnosis of endometrialadenocarcinoma,further determine endometrial adenocarcinoma,can be as a referenceindicator of endometrial adenocarcinoma prognosis.
