Objective:To establish and evaluate the Dexamethasone-induced osteoporosis model with type of deficiency of Kidney-Yin.Methods:45female Kunming mice were divided randomly into3groups, control group, model group and drug treatment group,15mice per group. The mice in model group and drug treatment group were given Dexamethasone Sodium Phosphate (DXM) by intramuscular injecting, whereas same amount of saline were given to the mice in control group by the same procedure. At the some time, the mice in drug treatment group were administered orally with Liuwei Dihuang pills suspension. The symptoms and signs of mice in each group were observed dynamically using quantitative score of symptoms and signs (QSSS). And the level of Adrenocorticotropic Hormone (ACTH), Thyroid Stimulating Hormone (TSH), estradiol (E2), cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP) were detected by ELISA to evaluate the condition of deficiency of Kidney-Yin. Bone mineral density (BMD) of proximal femurs and lumbar vertebrae2of mice in each group were tested dynamically. And morphological changes in lumbar vertebral were observed to evaluate osteoporosis.Results:(DQSSS:mice in model group appeared obvious kidney-Yin deficiency symptoms and signs, including hair dry, restlessness, easy excited, hard stool, urine yellow, and come to stable after given Dexamethasone Sodium Phosphate4weeks. At6weeks, compared with control group, the QSSS in model group were increased significantly (P<0.01). Compared with model group, the QSSS in drug treatment group were decreased significantly (P<0.01). There was no difference between control group and drug treatment group (P>0.05).②Hematology indices:At6weeks, compared with control group, the level of serum ACTH, TSH and cAMP/cGMP in model group mice were increased significantly (P<0.01, P<0.01, P<0.05), while E2decreased (P<0.01). Compared with model group, the level of serum E2in drug treatment group mice were increased significantly (P<0.01), while there was no difference between the level of ACTH, TSH and cAMP/cGMP (P>0.05, P>0.05, P>0.05). Compared with control group, the level of serum ACTH, TSH in drug treatment group mice were increased significantly (P<0.05, P<0.01), while there was no difference between the level of E2and cAMP/cGMP (P>0.05, P>0.05).③BMD:mice in model group appeared decreasing BMD of proximal femurs and lumbar vertebrae2, and come to stable after given Dexamethasone Sodium Phosphate4weeks. At6weeks, compared with control group, the BMD of proximal femurs and lumbar vertebrae2in model group were decreased significantly (P<0.01, P<0.05). Compared with model group, the BMD of proximal femurs and lumbar vertebrae2in drug treatment group were increased significantly (P<0.01, P<0.05). There was no difference between control group and drug treatment group (P>0.05, P>0.05).④Histomorphology:in model group, we could find significantly reduced and thinning trabeculae, disordered trabeculae, expanding medullary cavity and slight fractures. While in drug treatment group, it showed slightly disordered arrangement of trabeculae, slightly thinning of trabeculae, mild expanding medullary cavity and none obvious slight fractures, which is similar to control group.Conclusion:the model of osteoporosis with type of deficiency of Kidney-Yin could be established by Dexamethasone Sodium Phosphate intramuscular injection. Since had less intervenient factors, it is may be a stable and reliable modeling method for integration of disease and syndrome in TCM in the future.
