Objective: To explore the expression of the serine/threonine kinaseAKT/PKB and hexokinase2(HK2) in laryngeal squamous cell carcinoma andpara-carcinoma tissues and to study the relationship between them and allkinds of pathological factors of laryngeal carcinoma. Further, we investigatethe role and indication of PI3K-AKT pathway in regulating glycolysis duringthe occurrence, development, metastasis of laryngeal squamous cellcarcinoma in order to provide theoretical base for making treatment plan.Methods:30cases were chosen from the Second Hospital of HebeiMedical University from2012/2to2013/5which clinical data were completeand were confirmed by pathological examination before operation. Of the30patients,29were male and1was female, whose age range was from49to72and the average age was61.5±8.7yeas old. The patients had received neitherradiotherapy nor chemotherapy before operation who had no hyperthyroidismor hypothyroidism. We took the laryngeal squamous cell carcinoma andpara-carcinoma tissues as the objects of the research. The para-carcinomatissues were gained from the edge of the tumor which distance exceeded0.5cm (confirmed to be normal laryngeal tissues by pathology). The specimenwas put into the10%formalin fixative immediately after surgery,conventional paraffin-embeded, sectioned by5μm thick slice, deparaffinagedby xylene in convention, dehydrated by alcohol in gradient, removedendogenous peroxidase. The expression of AKT and HK2in carcinoma tissuesand para-carcinoma tissues was detected by immunohistochemicalStreptavidian-perosidase method (SP method). Then the slice were stainedwith DAB, hematoxylin stain, dehydrated, transparented and mounted by gum.All experimental data were analyzed by SPSS17.0statistical software whichP<0.05is statistical. The relativity between the protein positive expression and clinic pathological parameters was analyzed by Chi-square test, and usingSpearman’s Rank Correlation Analysis to test the correlation of AKT and HK2protein expression.Results:1The expression condition of AKT protein in laryngealsquamous cell carcinoma: the positive rate of AKT protein expression incarcinoma tissues and para-carcinoma tissues was66.67%and20.00%,respectively, and the difference was statistical significance (P<0.05). Thepositive rate of AKT protein expression in well differentiated laryngealsquamous cell carcinoma was50.00%and the positive expression rate inmoderate-poorly differentiated laryngeal squamous cell carcinoma was91.67%, and the difference was statistical significance (P<0.05). The positiveexpression rate of AKT in lymphatic metastasis group was90.91%andpositive expression rate was52.63%in no lymphatic metastasis group, and thedifference was statistical significance (P<0.05). But the positive expressionrate of AKT was not related to patient age, primary tumor position andclinical stage (P>0.05).2The expression condition of HK2protein in laryngeal squamous cellcarcinoma: the positive rate of HK2protein expression in carcinoma tissuesand para-carcinoma tissues was50.00%and13.33%, respectively, and thedifference was statistical significance (P<0.05). The positive rate of HK2protein expression in well differentiated laryngeal squamous cell carcinomawas33.33%and the positive expression rate in moderate-poorly differentiatedlaryngeal squamous cell carcinoma was75.00%, and the difference wasstatistical significance (P<0.05). The positive expression rate of AKT inlymphatic metastasis group was81.82%and positive expression rate was31.58%in no lymphatic metastasis group, and the difference was statisticalsignificance (P<0.05). But the positive expression rate of HK2was notrelated to patient age, primary tumor position and clinical stage (P>0.05).3The correlation of the expression of AKT and HK2protein: bySpearman’s Rank Correlation Analysis, data showed the expression of AKTwas positive related with the expression of HK2(P=0.019,r=0.424). Conclusions:1The expression of AKT and HK2protein in laryngealsquamous cell carcinoma was obviously higher than that in para-carcinomatissues.2The positive expression of AKT and HK2protein was related topathological differentiation degree and lymphatic metastasis, but was notrelated to patient age, primary tumor position and clinical stage.3The expression of AKT was positive related with HK2in laryngealsquamous cell carcinoma. The process of PI3K-AKT pathway positivelyregulating HK2advances glycolysis and inhibits cell apoptosis, which canpromote tumor’s occurrence and development and provide molecular biologyevidence for the clinical diagnosis, treatment, and estimating prognosis.
