Objective: As the rise of the morbidity and mortality of sepsis, it getsmore and more attention of people. Also caused a l-umber scholars to researchand discuss it’s pathogenesis in quite a long time. Although, so far it’spathogenesis is not yet fully understood., what is certain is that sepsis iscaused by infection of excessive inflammatory reaction, which leads to thewhole body system, organ and tissue function disorder and failure, resulting indeath. Early sepsis in immune system development can be characterized byimmune cells of amplification, enhanced immune function, can also be earlyimmunosuppressive phenomena directly.They Can even exist at the sametime, and influence each other, But the late sepsis is characteristiced byimmunosuppression. The extreme exhaustion of immune function increasedsecondary chance of infection, so as to increase the body’s infection. For theregulation of immune function in patients with sepsis has always been one ofthe important methods for the treatment of sepsis. Rhubarb can diarrhea,prevent endotoxin translocation and reduce the function of the inflammatoryresponse; Ulinastatin (UTI) can adjust immunity, enhance immune function,etc. Their treatments of sepsis each have certain curative effect,but thecombined application of rhubarb and ulinastatin to adjust the immune sepsispatients reported less. This experiment will be drug treatment in mice whichwere performedt artificial caecal ligation plus puncture(CLP). Through thechanges of T-lymphocyte subsets(CD3+、CD4+, CD8+, CD4+/CD8+) of ratsafter treatment to analysis and discussi UTI joint of rhubarb on the immunefunction of sepsis in rats, Contrast the treatment efficacy of each drug.Methods:120healthy adult SD rats (Sprague wrote-Dawley), weight210~250g, are divided into five groups according to the random number.Nornal control group, CLP model group, Rhubarb treatment group, UTI treatment group, Rhubarb combimed with UTI treatment group. Each group of24, male and female unlimited. Nornal control group immediately abdominalclosure after find the cecun. CLP model group adopted the melthod ofliterature, made the rats with abdominal cavity infected and subsequentlyappeared sepsis. Rhubarb treatment group will be drenched with rhubarb afterCLP (once every12hours). UTI treatment group will be Injected with UTIinto abdominal cavity after CLP(once every12hours). Rhubarb combimedwith UTI treatment group will be Injected with UTI and be drenched withrhubarb after CLP(once every12hours). Each group respectively in CLP after6h,12h,24h and36h four time points directly extract heart blood underanesthesia, each time point6in rats. By immunofluorescence labeling andflow cytometry instrument detection to analysis the content of each Tlymphocyte subsets in each group and to compare between groups.Results: Normal control group T lymphocytes of rats had no statisticaldifference compared with most of the literature reported data, P>0.05. Theexperimental rats without exception can be used as nomal control group CLP.Compared with the nomal control group, CD3+, CD4+,CD4+/CD8+of the6h ofthe CLP group showed a decline, had a statisitical difference, P<0.05. Withtime delay, the downward trend of CD3+, CD4+, CD4+/CD8+were more andmore obvious,showed that the immune suppression was appeared at thebeginning of the sepsis. Compared the Rhubarb treatment group with the CLP,the CD3+、CD4+、CD4+/CD8+of6h,12h,24h had no significant change. But at36h of the rhubarb treatment group, the above indexes appeared rising trend.The degree of immunosuppression is eased at the later stage. Compared theUTI reatment group with the CLP, immuno suppression did not appear at thebeginning of sepsis, the immuno function of rats was enhanced at thebeginning.And along with the time extension, CD3+, CD4+, CD4+/CD8+ratioincreased significantly, the immuno function improved significantly, themortality of rats significantly decreased. The results of Rhubarb combimedwith UTI treatment group was similar with UTI treatment group.CD3+, CD4+,CD4+/CD8+had a early rise. As the extension of time, the immune regulating function improved significantly, especially24h and36h.Conclusion: Rhubarb has diarrhea, inhibit inflammatory effects, makingit easier for the inflammatory response in rats relieve and under control. It alsohas a certain therapeutic effect to prevent immunosuppression in late sepsis.Ulinastatin has a strong immune regulation ability, and can be displayed on theearly pathogenesis of sepsis. The immune function improved obvious. CD3+,CD4+, CD4+/CD8+increased significantly, the prognosis can improved greatly.Rhubarb and UTI respectively have certain adjustment on the immune andinflammatory response, they have synergistic effect and have better regulationon immune function in rats. Both of rhubarb, UTI, UTI combined withrhubarb have no obvious effect on the CD8+...
