Loquat leaves is a traditional Chinese medicine.It’s a long time that the Loquat leaves is used as a medicine. Now the results of the Pharmacological studies declare that the Loquat leaves also can anti-tumor, anti-infective, contra-virus, Hypoglycemic,reducing blood fat, hepatoprotection,cholaneresis and regulate immune function. In this research,I will determine the content of triterpenic acid, the volatile component. Inorganic elements in loquat from FuJian, then get the finger printing of the effective component. Except these I will study on the protein components. It’s expect that the results of this research will provide a scientific basis to the rational use of the loquat leaves and the intrinsic quality assessment of the traditional Chinese medicine.The loquat in this reserch got from the GAP production base in PuTian city of Fujian and other areas. I have determined the content of water in the leaves and the extractum in water or alcohol with the method are offerd by Pharmacopoeia.Determining the content of oleanolic acid and ursolic acid by HPLC. Extraction volatile components of loquat with Supercritical CO2Extraction, then using the GC-MS determine The content of volatile components. Obtaining the finger printing of the effective component by HPLC.Extracting the protein in loquat by TCA acetone,after measure the contenr with BCA method, study the protein bands with SDS-PAGE gel electrophoresis. Determining the contents of inorganic elements in loquat which from different areas by ICP-MS.The data from the experimental shows:1、extractum in water or alcohol of loquat are different among the varieries. Three varieties of loquat from Fujian have higher quality,they are jiefangzhong、changhong3and zaozhong6.But Hunan Baisha, Ruantiaobaisha, Jiajiao have lower content of oleanolic acid and ursolic acid.If the drying temperature exceed100℃, the extractum will decline sharply.In addition,the content of loquat on the tree is higher than the leaves had followen.2、The content of oleanolic acid and ursolic acid in loquat are different among23varieries.The highest is zhiefangzhong,about two times to the HuNan baisha which is the lowest one.The highest content of both is zhefangzhong.Jiajiao have the lowest content of oleanolic acid and the HuNan baisha has the lowwest ursolic acid.Comparing to the samples from the GAP production base,the total content of oleanolic acid and ursolic acid are higer than the wild species.3、The results of GC-MS show that the main volatile components in loquat are alkenes and enols. And the volatile substances in mature leaves and the tender leaves are singnificant differentThe main ingredient are7-stigmast en-3-ol, betulin,5-ergot en-3-ol.4、The degree of similarity of grafting and shishengmiao is over0.9,and the fingerprintd and the kinds of chemical components of two samples are basically the same.But the degree of similarity in four wild samples are no more than others.5、The protein bands have apparent different at70-90KD、70-50KD、20KD,from this result we can know that the differents among the different loquat are higher the same variety.6、From the results of ICP-MS, we can know that the content of Ca, Fe, Mg, Al, K are higher than other elements, Cu、Mn are higher than the other heavy metal elements.The content of these elements in loquat have significant different. So the soil will influence the quality of the same species of loquat.Analyzing the results of the experimental we can kown that the content of triterpenic acid, the volatile component are influence by the special, place of origin, age, processing methods. the quality of different loquat are diversity in the same cultural environment.Beside this the different soil and growing conditions will make the diversity in the same kinds of loquat.But the harvest season and the reproduction have no significant effect on the chemical quality of loquat.This provide a scientific basisto expand resources and make full use of traditional Chinese medicine. Analyzing the results of the SDS-PAGE electrophoresis.It shows Protein components of different loquat are diversity. Weather this result is related to the planting environment,then affect the chemical composition is need a further and depth study.This research project provide experimental basis for the studying that the correlations between the chemical composition and proteins of loquat.
