Objectives:1.To observe the effects of troxerutin intraperitoneal injectionof long dorsal random skin flaps survival in rats.2.Explore its possiblemechanism, in order to deepen the knowledge of ischemic random flap,toimprove the skin flap ischemia in practice,provide a new method to promote flap survival.Methods:Select24female SD rats,weighing about200g,were randomly divided into experimental group and control group.By intraperitoneal injection of2%pentobarbital sodium (20mg/kg) under general anesthesia,the rats were fixed,then shaved the surgery area on their backs.The random flaps were designed a pedicle in the head side about the size of7cm×2cm on central back of the rats,then disinfecting with the iodophordisinfectant and drape,along the design lines cut the skin and raised from the panniculus carnosus,after hemostasis,the flaps were sutured back into their original places with3-0silk suture.Rats were given intraperitoneal injection with troxerutin(20mg/kg) in experimental group,once a day,for seven consecutive days,the control group rats received the same volume of saline by the same way.The changes of flaps were observed after operation.7days later,all rats were executed,the survival area of the flap and the survival percentage calculation,cut the middle flap tissue from the two groups, immunohistochemical method observation of microvascular number under light microscope,in three sections,the skin flaps were cut for histopathology examination and to test superoxide dismutase(SOD) activity and malondialdehyde values(MDA).Results:The percentage of skin flap survival was higer in troxerutin group than that in control group(78.90±4.90and56.41±6.54, respectively, P<0.05).Observation of pathology:Each organization in experimental group,necrosis and inflammatory cell infiltration was lighter than that in control group.In the troxerutin group the microvascular number was larger than that in control group (15.35±1.09and12.81±0.83, respectively, P<0.05).In experimental group the near、middle and distal segment,the MDA values was lower (P<0.05),while,the SOD activity was apparently higher (P<0.05)than that in control group.Conclusions:Troxerutin can promote flap survival by reducing tissue edema and inflammatory cell infiltration,by increasing microvascular number to improve micro-circulation of flaps,and by enhancing SOD activity and reduce the level of MDA.
