Anthocyanins and polyphenols are both natural antioxidants. It has previously indicated that anthocyanins and polyphenols effectively eliminate free radicals. In addition, they also play important roles in tumor, cancer, cardiovascular diseases and eye diseases. In this thesis, eight kind of plant extracts have been used for the visual research, they were four kind of anthocyanins include cyanidin-3-O-glucoside (C3G), bilberry extract, purple rice extract, mulberry extract and four kind of natural tea products involve epigallocatechin-3-gallate, tea extract, L-theanine, caffeine. There are mainly three steps in this study, the first is rhodopsin recovery rate determination and the second is retinal ganglion cell viability determination, the last is to design a formulation about diseases eye protecting and treatment. Measuring different rhodopsin recovery rate to test the Synergies of C3G and EGCG on rhodopsin recovery rate which complete depend on11-cis-retinal (11-cR) and free bullfrog opsin membranes(OM) linking into rhodopsin. At the same time, study the changes of rhodopsin decomposition rate following EGCG and C3G through calculating the content ratio of rhodopsin under different illumination time. In this experiment,11-CR is obtained by high-speed countercurrent chromatography in a final concentration97.5%after light produced all-trans-retinal(all-tR) isomerization. Experimental results show that, C3G and EGCG can significantly improve the rate of recovery of rhodopsin. When the C:E=2:1, the recovery rate increased2.4-fold compared with the blank group. In addition, the data shows that EGCG can significantly facilitate C3G accelerate the recovery rate of rhodopsin. The rhodopsin retention study results show that C3G and EGCG have a protective effect on the degradation of rhodopsin.Different concentrations of C3G, bilberry extract, purple rice extract, mulberry extract, EGCG, L-theanine, tea extract and caffeine have been used for detect both protection and repair capacity to hydrogen peroxide (H2O2) and UV induced retinal ganglion cells (RGC-5) oxidative stress. Our study results show that:C3G, bilberry extract, EGCG and tea extract have good protection in the damage caused by H2O2and UV. At present research, these natural products have no significant repair efficacy for damaged RGC-5cells. L-theanine and caffeine have significant role in promoting cell growth. The cell proliferation rate increases gradually as the concentration increased. In addition, both of them can well protect and repair damaged cells. Especially, L-theanine has a significant role in the protection and restoration to UV-induced cell damage, the highest rate of protection and repair rates were47.1%and35.1%.Measured the following six data to evaluate visual protection and repair capacity of each formulation. The six data were:rhodopsin recovery rate, cell growth proliferation rate, cell viability of protection group by H2O2injury, cell viability of protection group by UV damage, cell viability of repair group by H2O2damage, cell viability of repair group by UV damage. Finally determined the best formula is:56.6%L-theanine, caffeine19%, EGCG5.4%, Bilberry Extract19%.
