Cordyceps sinensis (Berk.) Saccl. is a famous fungus which widely used on Chinese herb.Considering the scarce yield of natural Cordyceps sinensis, it’s very important to study onproducing Cordyceps sinensis mycelium and some functional ingredients by submerged culture.This research focused on submerged culture technology, extraction technology and biologicalactivity of polysaccharide of Cordyceps sinensis.Results showed that it could get solid culture condition of excellula Cordyceps sinensis andchoose the best culture medium and condition by mycelium wet weight and polysaccharideefficiency measure. The best liquid seed culture medium was consisted3%glucose,1%peptone,0.4%KH2PO4and0.2%MgSO4. The best culture conditions were10%inoculation,140r/min,24℃and pH7.0.The wet weight and polysaccharide efficiency of Cordyceps sinensis wererespectively102g/L and7.45g/L with optimized culture conditions.The research optimized of extraction, separation and purification conditions for advancedefficiency of excellular polysaccharide of Cordyceps sinensis. The results showed that thebest condition of deposition of excellula polysaccharide of Cordyceps sinensis were-15℃,24hrs and pH7.0with3times ethanol. The best condition of deposition of incellulapolysaccharide of Cordyceps sinensis were85℃,2hrs and pH7.0with3times water. It wasused Sevag method to separate protein.The test demonstrated that polysaccharide of Cordyceps sinensis can improve animalimmunity. Different effects were shown in different doses about mice experiment. Incellulapolysaccharide of Cordyceps sinensis with protein improved weight of liver and spleen,proliferation efficiency, phagocytosis index, IL-2and IFN-γ significantly. Compared with thecontrol group, they increased1.22,1.32,1.78,1.27,1.29and1.27times respectively. Butthere was no significant difference between the control group and incellula polysaccharide ofCordyceps sinensis without crude polysaccharide group that separated by SephadexG-100method. The results confirmed that Cordyceps sinensis with protein improved immunityability more significantly.
