Objective: The underlying mechanisms of Diabetes Mellitus (DM) increasing the riskof Coronary Heart Disease(CHD) so far has not fully established. The purpose of thisproject is to discuss the relationship between AKT2gene polymorphisms with theoccuring of Type2Diabetes Mellitus (T2DM)and coronary heart disease in thepopulation of north China.Methods: A total of136hospitalized patients (72males,64females) were selectedfrom the department of Cardiovascular Medicine and Endocrinology which belong tothe Second Hospital of Jilin University from October2013to March2014. They weredivided into3groups:58in Coronary Heart Disease group (CHD);58in CoronaryHeart Disease with Diabetes group (CHD+DM);20in control group (Controlgroup).After extracting the genomic DNA, and through the PCR amplification of SNPloci from clips, applicate the SNP mass spectrometry analysis AKT2gene20activeloci genotype frequency, and choosing different types of peak for DNA sequencing.After the calculation of genotype frequency and allele frequency, by the Personchi-square analysis the differences on each allele site of disease group and controlgroup, then looking for the association between the sites and disease.Results: According to the results of study, there is statistically significant differencebetween Diabetes with CHD and simple CHD group in the genotype of AKT2rs3730051(p<0.05).After statistical analysis, The mutant allele G of AKT2rs892119and the mutant allele A of rs3730051carried a higher risk of Coronary Heart Disease; and there is also statistically significant between the case and control group on thedifference of rs8100018(p<0.05),the patients with genotype GG significantlyincreased the risk of T2DM.Conclusion:1、There is an association on AKT2gene loci rs8100018for Type2Diabetes Mellitus in people of northern China.2、There is an association on mutant alleles G and A of AKT2rs892119and rs3730051of increasing the incidence of Coronary Heart Disease in thepopulation of northern China.Therefore, it can be verified that AKT2gene is the susceptibility gene of Type2Diabetes because of the rs8100018sites. The mutation of AKT2gene locusrs892119and rs3730051can increase the incidence of Coronary Heart Disease.
