| Objective:Doxorubicin is an efficient anticancer drug in the clinicalcancer treatment,but its adverse effect-cardiaoxicity limitits use.It caninduce late cardiopathy. In this test,we compare the results to constructthe proper and simple models.Methods: In this test, we use Wistar male rats, they were dividedinto3groups(n=10) at random injected with DOX and the correspondingcontrol groups: in the test group, eachrat was injected with DOX via veinof3mg/kg once a week, and the each control rat was injected with theequal volume of saline via tail vein at the same time, both for six weeksin total, they are killed at the sixth or seventh or eighth week, during thetest, the weight, mortality and the body weight are observed. The cardiacfunction was tested, and the serum TnI concentration was measured, wealso detect the morphological and ultrastructural transformation ofmyocardial mulsle,and analyzed the results statistically.Results:compared with the control groups, the body weight of thetest group is inhibited. A compare with a, B compare with b, C comparewith C, the body weight decrease significantly, P<0.05.A compare withB,P<0.05.B compare with C,P<005.Mortality in A,B,C is10%,30%and60%respectively.In the test, the hemodynamic results showed that there no significantdifference between A and a, while there are significant difference betweenB and b, as well as C and c. Interestingly, the dynamic data in B havesignificant decrease comparing with A and significant increase comparingwith C.A compare with a, there are suggestible differences in the TnI level,but no differences in the BNP level. But comparing group B or group Cwith the corresponding control group, the significant difference exist both in TnI level and BNP level.Test group A, B and C compare with the respective control group,theHW/BW rate decrease significantly, and A compare with B, P<0.05, Bcompare with C, P<0.05, but there are no difference between A and B.The morphological and ultrastructural suggest that the DOX group isdamaged severesly to different extend, in A, the damage is the mostslightly.Conclusion:In the test, the test grouprat was injected with DOX viavein of3mg/kg once a week. A compare with a, there are no suggestibledifferences in the TnI level and the morphological structure. B comparewith b, the results showed that the contracting rate and the diastole rate isobviously discend. The TnI concentration ascend. The morphological andultrastructural suggest that the DOX group is damaged severesly, theconstruction of the myofibers are disordered. In the C group,8rats diedin the end. So, group B is the ideal model imitating the progress ofinjecting drugs clinically... |
