Objective: To determine the effects of silibinin(SB) on proliferationinhibition and apoptosis of human bladder cancer T24and5637cells,and to study the possible mechanism.Methods: Human bladder cancer5637and T24cells were culturedin cells media and the logarithmic growth phase of the5637and T24cellswere divided into control group and25、50、100、200'400μmol·L-1SBgroup. The effects of SB on the proliferation inhibition of T24and5637cells were evaluated by the MTT assay; inverted phase contrastmicroscope was used to observe the invasion inhibition ability of T24and5637cells after treated with different contentations of SB; Themorphological changes of T24and5637cells after treatment wereobserved by DAPI staining; Ability of apoptosis was detected andevaluated by flow cytometer.Results: MTT results show that compared with the control group,the cell viability rates in T24and5637groups were lower than that incontrol group (p<0.05). After treated with different concentrations of SB,the migration of T24and5637cells were significantly inhibited. DAPIstaining and flow cytometric results revealed that cell apoptosis wasobserved after different concentrations of SB, the apoptotic rates in SB groups were higher than that in control group(p<0.05).Conclusion: SB could inhibit the proliferation and invasion ofhuman bladder cancers T24and5637through induction the apoptosis ofcells.
