Objective: To detect the expression of TACC3 protein in bladder urothelial carcinoma and study the influence of its expression level to proliferation and apoptosis in T24 cells.Methods: The expression of TACC3 protein in 35 cases of bladder urothelial carcinoma and 35 cases of normal mucous was detected by western blot and the clinical pathological features were analyzed; Through in vitro studies, we design and construct several sh RNA expression plasmid and screen the most effective target through q PCR. Then we use cytology transfection technique to interference TACC3 level in bladder cancer cells, western blotting to detect the interference effect of RNA, through MTT to measure the growth inhibition rate of T24 and Flow cytometry to verify its apoptosis rate.Result: 1. The relative grey levels of bladder urothelial carcinoma tissues and normal mucous respectively are 0.62±0.10 and 0.29±0.14. The difference was statistically significant. While the expression level of TACC3 between patients’ sex, age,tumor size and number had no statistical significance differences;And statistically significant difference were found between pathological staging,tumor degree and smoking(P<0.05). 2. We successfully built 4 TACC3-sh RNA interference plasmids,and use Hela cells to select a most efficient TACC3-sh RNA.Its interference effect as high as 90.5%. 3. Result of MTT showed the growth inhibition rate of T24-TACC3-sh RNA was 62.24%(48h). Flow cytometry showed its apoptosis rate was 39.57%(48h).Conclusion: 1. TACC3 is highly expressed in bladder urothelial carcinoma. Normal bladder mucosa epithelial weakly positive expression. 2. The expression level of TACC3 was positively related with pathological stage,tumor differentiation degree and smoking.3. TACC3-sh RNA can inhibit the proliferation of T24 bladder cancer cells, and promote its apoptosis. 4. The result suggest that TACC3 may play an important role in the development and progression of bladder urothelial cancer.It would be an important biomarker candidate,and providing a new way for the diagnosis and treatment of bladder urothelial cancer.
