Objective: Normal cervical tissues, CIN III tissues and cervicalcancer tissues are compared for spectral analysis using Ramanspectroscopy. We summarize the respective characteristic peaks,and explore whether the Raman spectroscopy could be used for theclinical screening of cervical cancer and cervical cancer lesionbefore or not.Materials and methods:Tissue samples were taken from63casespatients of surgical treatment during April2013-November2013(Department of gynecology, Jilin University First Hospital). Amongthem,33cases were cervical squamous cell carcinoma of theorganization,10cases were cervical intraepithelial neoplasia IIIgroup as the experimental group. normal cervical tissues and CINIII tissues and cervical cancer tissues were analyzed using Ramanspectroscopy with regard to spectroscopy analysis, then results ofRaman spectroscopy of different groups were analyzed.Results:1.Raman spectra in different tissues possesses differentcharacteristic peaks position. The specific performance are: maincharacteristic peak in the Raman spectra of normal cervical tissue isat640,853,928,1005,1242,1450,1660cm-1; main characteristicpeak in the Raman spectra of CIN III tissues is at853,931,1003,1242,1341,1447,1657cm-1; main characteristic peak in the Ramanspectra of cervical cancer tissues is at642,853,1003,1345,1443,1657cm-1. 2. The same Raman spectra characteristic peak of different tissueshas different relative strength: Nucleic acid as the maincharacteristic peak is1345cm-1, the nucleic acid content is differentand ordered by cervical cancer> CIN III> normal cervical tissues(P <0.01); Protein as the main characteristic peak is at1003cm-1,protein content differs dramatically and ordered by cervical cancer> CIN III> normal cervical tissues (P <0.01); Lipid as the maincharacteristic peak is at1443cm-1, lipid content of normal tissuesis much more than the cervical intraepithelial neoplasia III tissuesand cervical cancer, however, there is no significant differencebetween them (P>0.05).Conclusion:1The characteristic peaks location of Ramanspectroscopy of normal cervical tissues, CIN III tissues and cervicalcancer tissuestissues is different.2The same characteristic peakrelative strength of Raman spectra in normal cervical tissues, CINIII tissues and cervical cancer tissuesis different.3Ramanspectroscopy is expected to become the measuring method forscreening cervical cancer and cervical cancer soon.
