Effects Of Gan Cao Xie Xin Decoction On The IL-8、I-κb In Rats With Experimental Ulcerative Colitis

1. Background and Objective:In order to observe the influence of Gan Cao Xie Xin Decoction on the Inflammatory cytokines in rats with experimental ulcerative colitis,and identify its underlying mechanism of action.2. Method:Totally90healthy male Wistar rats were divided randomly into nine groups:normal group,model group,sasp group,Xie Xin Decoction High dose gavage group,Xie Xin Decoction Medium dose gavage group,Xie Xin Decoction low dose gavage group,Xie Xin Decoction High dose clyster group,Xie Xin Decoction Medium dose clyster group,Xie Xin Decoction low dose clyster group,Ten rats in each group.We prepared the models by means of the2,4-Dinitrochlorobenzene/Glacial acetic acid enema except the normal group, and the experimental group were given gavage or clyster normal saline, SASP,Gan Cao Xie Xin Decoction,respectively.The medication was from the3rd day to the17th day after the model was established. At the end of the experiment,the disease activity index (DAI) was evaluated.The rats were anesthetized by the chloral hydrate, and dissected to get the colon tissue and blood.The colonic macroscopic damage index (CMDI)and histopathology were evaluated,the serum levels of interleukin-8in the way of enzyme-linked immunosorbant assay(ELISA),and the level of1-Kb expression in the colon by means of immunohistochemical.3. Result:The score of DAI and CMDI were significantly increased in the model group as compared with that in the normal group(P<0.05),and compared with the model group,DAI and CMDI of the treatment groups declined significantly(P<0.05),but no significant differences were found in the treatment groups(P>0.05).Comparing with the normal group,The serum levels of IL-8was increased significantly in the model group(P<0.01),the serum levels of IL-8was decreased significantly in the treatment groups as compared with the model group(P<0.01),but no significant differences were found in the SASP group and Xie Xin Decoction treatment groups(P>0.05).Xie Xin Decoction Medium and low dose clyster groups were better than other Xie Xin Decoction treatment groups,and significant differences were found(P<0.05). Comparing with the normal group,The the level of I-Kb expression in the colon was increased significantly in the model group(P<0.01),the the level of I-Kb expression in the colon was decreased significantly in the treatment groups as compared with the model group(P<0.01),Xie Xin Decoction Medium and low dose clyster groups were better than other Xie Xin Decoction treatment groups and SASP group,significant differences were found(P<0.05).4. Conclusion: Gan Cao Xie Xin Decoction can reduce IL-8,and Truncated the activation pathway of NF-k B,and promote the integration of I-k B and NF-KB,then reduce the activity of NF-KB,and alleviate intestinal inflammation.Thus it played a role in treatment of UC.
