Objective:Using proteomics methods to study on protein fingerprints of IgA nephropathy Qi and yin Deficiency syndrome, looking for specific protein expression profiles of IgA nephropathy with Qi and yin Deficiency syndrome, explore the serum proteomics marker of IgA nephropathy with Qi and yin Deficiency syndrome from proteomic level.Method:Collecting fasting serum of patients aged from18to60years old in the morning from our hospital patients from2012-2to2013-2the diagnosis was IgA nephropathy, Combining with the classification criteria of TCM syndrome for chronic primary glomerular disease which is adopted by second national nephropathy thematic colloquium,as well as the international blood stasis diagnostic criteria, formulated by international conference on research blood stasis syndrome in1988,with a combination of laboratory coagulation indicators, normal fasting serum samples were collected at the same time. Using proteomics methods (SELDI-TOF-MS technology and metal ion chelating protein chip) and statistical methods, at the same time using Biomarker PatternsTM Software (BPS), to analyze serum protein fingerprint of each group. By further screening and establishing the decision tree and ROC curve, and the test diagnostic model, Looking for specific serum protein expression of IgA nephropathy with qi and Yin deficiency syndrome.Result:There were14cases of qi and Yin deficiency syndrome and14cases of blood stasis syndrome and14cases of normal fasting serum samples. Experimental results were as follows:1th, IgA nephropathy with qi and Yin deficiency syndrome compared with normal group, it was found that22protein peaks were significantly different (P<0.05),which11protein peaks were high expression in the group of qi and Yin deficiency syndrome (including M/Z1972.32),11protein peaks were lower expression in the group of qi and Yin deficiency syndrome.2th, It was found that29protein peaks (no including M/Z1972.32) were significantly different between the IgA nephropathy patients with blood stasis syndrome and normal group(P<0.05),which23protein peaks were high level expression,6protein peaks were lower expression in the group of blood stasis syndrome.3th, Five protein peaks (M/Z:4987.92、7595.90、8272.85、23045.1、5511.47)between three groups were different, significant was Meaningful (P<0.05).4th, IgA nephropathy patients with qi and Yin Deficiency group compared with the blood stasis syndrome were found in19different protein peaks and were statistically significant (P<0.05), which two protein peaks were high expression (M/Z:1972.32.6485.68). Based on the above analysis and Biomarker Patterns TM Software, five markers (M/Z:1972.32>6759.43.4380.12,4689.68.2893.68) were found, the sensitivity of the decision tree model was85.71%and the specificity was89.29%.Conclusion:M/Z1972.32proteins may be the specific serum protein marker of IgA nephropathy with Qi and yin deficiency syndrome.
