Objctive:Prediabetes should be viewed as risk factor for cardiovascular,and the mechanism is unknown.We suppose soluble receptor for advanced glycation end products(sRAGE),endogenous secretory receptor for advanced glycation end products(esRAGE) and oxidative stress had changed in prediabetes.This study investigates sRAGE,esRAGE and oxidative stress in the normal glucose tolerlance(NGT),prediabetes(Pre-DM) and newly diagnosed diabetes(DM),and explored the relationship between sRAGE,esRAGE and oxidative stress in prediabetes.We aimed to provide evidence for the pathogenesis of elevated risk of cardiovascular events with prediabetes.Method:All subjects underwent a75-g oral glucose tolerance test (OGTT) and insulin releasing test.The subjects were stratified into three groups according to the plasma glucose.Fasting plasma was stored to examined sRAGE,esRAGE,8-isoprostaglandin F2α(8-iso-PGF2α),total antioxidant capacity(TAOC) superoxide dismutase(SOD) and malondialdehyde(MDA).Result:Plasma sRAGE,esRAGE and SOD were significantly lower in Pre-DM and DM compared with NGT,plasma8-iso-PGF2a and MDA were significantly higher in Pre-DM and DM compared with NGT,the difference was statistically significant(p<0.05).Plasma TAOC was significantly higher in Pre-DM and NGT compared with DM, the difference was statistically significant(P<0.05).In Pre-DM,sRAGE had a significantly positive correlation with esRAGE (r=0.382,p=0.007),had a significantly negative correlation with HOMA-IR, MDA and8-iso-PGF2α(r=-0.314,-0.313,-0.34, p=0.028,0.028,0.016, respectively). esRAGE had a significantly positive correlation with sRAGE and TAOC (r=0.382,0.598, p=0.007,<0.001),had a significantly negative correlation with MDA(r=-0.397, p=0.005).By the use of multiple stepwise regression analyses,esRAGE,HOMA-IR and8-iso-PGF2a remained significant independently with sRAGE(R2=0.374);sRAGE and TAOC remained significant independently with esRAGE(R2=0.445).Conlusion:Plasma sRAGE,esRAGE and oxidative stress had changed in prediabetes,suggesting that sRAGE and esRAGE might play an important role in oxidative stress and antioxidative in vivo.
