As the fluorescence analysis technology is used in the field of life sciences, various of fluorescent probes which can specific label biomolecules are developed.As one of the important members, Indocyanine dye has the advantage of tunable absorption wavelength, high quantum yields, easily be modified, and good stability. This paper selected the heptamethine cyanine as parent and synthesized near-infrared fluorescent dyes that modified with carboxyl group and boronic acid group respectively. Then selected appropriate methods to purify them and did a preliminary exploration of the carboxyl group-modified dye’s application.The first chapter firstly summarized indocyanine dye’s application in bio-analysis, optical disk storage, the sensitive photographic and solar cells. Then summarized the progress of synthesizing of the trimethine,The pentamethine and the heptamethine indocyanine dyes.Thus determined the purpose and significance of the thesis topics.The second chapter described the routes and methods of synthesizing the carboxyl-modified fluorescent dye Dyel. Appropriate methods were taken to characterize the intermediates and Dyel. Symmetrical fluorescent dyes Dye2and Dye3which had similar structures were also synthesized in the same way.According to the different charge characteristics among the intermediates and Dyel, the third chapter introduced a new method which uses DEAE-cellulose anion exchange chromatography to purify the taget dye. The pre-experiment using symmetrical dyes Dye2and Dye3proved the separation degree. Comparing with the gel chromatography, this method has shown its advantage.In the fourth chapter, mouse monoclonal antibody CRP was labled by Dyel and CFTM647SE, respectively. The D/P showed that these two kinds of dyes have the similar practicality. The last chapter synthesized the designed boronic acid-modified dye. Purified the asymmetrical Dye4and symmetrical Dye5using gel permeation chromatography G-25and confirmed their strcture with’H NMR.
