Carbohydrate is extremely important to human beings , it is not only the food source for people to survive ,but also the important information material of lives. It is crucial to cell recognition, interaction, water and electrolyte transportation, generation and metastasis of cancer, immunization and immunosuppressive of organism, as well as the fertilization and cell agglutination and other biological processes. Therefore, it is very important to develop the high sensitivity, high selectivity recognition approchs.Organic boronic acid can be combined with carbohydrate, therefore, it can be used as a recognition group for carbohydrate, the saccharide sensor based on boronic acid has drawn extensively attention in recent years. Based on previous study, a fluorescence detection method was chosen in this paper, the boronic acid was combined with fluorescent materials to establish a fluorescent probe. When the probe combined with saccharides, the fluorescence property of the probe was changed, the fluorescence changes were measured to derive the sugar related information. For this, four new saccharide detection methods were designed in this paper, and the preliminary exploration was studied for each, this exploration will lay the experimental foundation of the further study. The thesis includes the following sections:1. The thiopheneboronic acid modified gold nanoparticles based saccharides probe (probeâ… ) was prepared ,the gold nanoparticles was synthesized by sodium citrate reduction method, then gold nanoparticles were modified by thiopheneboronic acid to achieve probeâ… ,and the influence factors were investigated to determine the optimal experimental conditions, at the optimum conditions, different type and different concentration of saccharides were added to determine the response sensitivity of probeâ… .2. The ultraviolet fluorescence reagent of benzeneboronic acid NAA-APBA (probeâ…¡) were synthesized by the reaction of EDC and NHS. The influence factors were investigated to determine the optimal experimental conditions, at the optimum conditions, different type and different concentration of saccharides were added to determine the response sensitivity of probeâ…¡.3. The mercaptoboronic acid modified gold nanoparticles (probeâ…¢) were synthesized with the thiol self-assembly and solid-phase methods. The influence factors were investigated to determine the optimal experimental conditions, at the optimum conditions, different type and different concentration of saccharides were added to determine the response sensitivity of probeâ…¢.4. Rhodanmine based fluorescence probe (probeâ…£) for saccharides was synthesized. The influence factors were investigated to determine the optimal experimental conditions, at the optimum conditions, different type and different concentration of saccharides were added to determine the response sensitivity of probeâ…£.The boronic acid based saccharides probes designed in this paper were characterized by simple to prepare , high sensitivity, easy to use and so on. The preparation method of probeâ… is relatively new ,and it was reported rarely at home and abroad. In pH7.3, close to physiological pH, probeâ… produced a high sensitivity to glucose, therefore probeâ… has the potential to be developed into a blood glucose sensor used in clinic, this research has laid a theoretical and experimental foundation for developing a new type of glucose detection device.
