Objective: To explore the expression of ICAM-1ã€NF-KBand its role of emergencyand development in SAP with HL。In order to provide further clinical material for HAPmicirculation.Methods: Forty male SD rats were randomly divided into4groups: SAP group(groupA),HLSAPgroup(group B),HL and sham operation group(group C),sham operationgroup(group D).The specific methods are follows:First,the rats were randomly dividedinto two groups(N=20),respectively feeding normal diet and high-fat diet (which consistof87.8%normal diet,10%lard,2%cholesterol and0.2%sodium cholate)for4weeks,eachgroup was randomly devided into two groups(n=10)again and used retrograde injection5%sodium taurocholate which induced sereve acute pancreatitis and only flipped to thepancreas and duodenum after opening the abdomen.Rats were sacrificed,and thematerials were gained at12hours after models were made.To measure the value ofamylase(AMS),triglyceride(TG), endothein-1(ET-1)in serum.Open zhe abdomen againto observe ascites,saponification spot and the alteration of pancreas in general. Thetissue of the pancreas were observed through light microscopy after HE staining and thepathological score were evaluated.ICAM-1and NF-kB in pancreatic tissue wereexamined by immunohistochemistry.Results:1.The concentration of triglyceride(TG):After the high-fat diet four fourweeks, the levels of TG were significantly higher then that before the high-fatdiet(P<0.01).The seum TG levels in group B were significantly elevated as comparedwith group Aand group D(P<0.01),.The seum TG levels in groupA wre higher then than of group C,but there was no statistical significance(p>0.05).2. The concentration ofamylase(AMS):The serum AMS levels in both A and B groups were significantlyelevated as compared with group C and group D(P<0.05), but, there was no different ingroup A and group B(p>0.05).3. The concentration of endothein-1(ET-1):The levels ofET-1both in group Aand group B were significantly higher then that og group CandD(P<0.01),in the sametime,group B were hingher then group A(p<0.05),there was nodifference with group C and group D(p>0.05).4. The general situation to the abdomenthe pathological score of the pancreas: It can be observed pancreasoedematous,engorgement, necrotizing,ascites in group A and group B after severeacute pancreatitis had been induced. the histological degree of pancreaticdamage were significantly higher in group Aand group B than that in sham operationgroup(p<0.01).The score of the edema and infiltration of inflammatory cell in bothgroup Aand group B has no difference(p>0.05),but the hemorrhage and necrosis scorein group B were significantly higher than that of group A(p<0.05).5.The result ofimmunohistochemistry:The positive cells population of ICAM-1and NF-kB inpancreatic tissue in both and group B were significantly higher then that of shamoperation group(p<0.01). The positive cells population of ICAM-1and NF-kB inpancreatic tissue in group B were significantly higher then that of group A(p<0.05).Theconcentration of TG and the positive cells population of ICAM-1and NF-kB werepositive correlation in group B,the correlation coefficient were0.946and0.923.Conclusions:1.An idea animal model of HL can be bulided by feeding high-fat diet for4weeks.A model of SAP complicated with hyperlipemia in rat might be developed byfeeding high-fat diet for4weeks and retrograde injection of5%sodium taurocholateinto the pancreatic duct.2. Hypertriglyceridemia intensifies the pathologic-histologicaldegree of pancreatic damage.3. Hypertriglyceridemia can improved ET-1rising morehigher in severe acute pancreatitis, the concentration of TG and ET-1were positivecorrelation.4. Hyperlipidemia may intensifies the expression of ICAM-1and NF-kB in rats with severe acute pancreatitis. |
