Objective: By observing the impact of acupuncture on the T2DM rat hypothalamicGLUT3and GLUT4gene expression and protein content; Further explore the GLUTsfamily in the hypothalamic insulin signal transduction in type2diabetic rats frommolecular level; At the same time explore influence of effector mechanisms in the Glutsfamily acupuncture treatment of type2diabetes; study the possible mechanism, toprovide a theoretical basis for clinical acupuncture treatment of T2DM.Methods:1.60SD mate rats, average weight about220g. The computer randomly generated10for blank control group, fed with normal diet for4weeks; the remaining with high fatdiet for4weeks, then manufactured model, and retained20qualified screening diabeticrats,, the computer randomly divided into2groups, the acupuncture group and themodel group respectively,10rats in each group. A total of30rats, divided into anacupuncture group, model group and blank control group, with10rats in each group.The rats of acupuncture group acupuncture the Yishu, Zusanli, Neiting,15minutes eachtime,1time per day for2weeks. The model group:like the acupuncture group, but notacupuncture and any other treatment. Blank group: like the acupuncture group, but notacupuncture and any other treatment, treatment course for2weeks too. In theexperimental process, each group had a rat death, Based on the nine rats in each group.2.Group intervention after2weeks, fasting overnight, in the next morning at8, groupsof SD rats with10%chloral hydrate (0.35ml/100g) intraperitoneal injection ofanesthesia, the left ventricular blood5ml, static separation of serum tested indexes. Theacupuncture group, model group and blank control group real-time sampling:guillotined,put in ice dish, rapidly isolated of the hypothalamus and quadriceps femoris,0.9%thephysiological saline flushed surface blood, Gauze sucking, liquid nitrogen preservation. Determination of glucose oxidase, the content of muscle glycogen; radioimmunoassayin peripheral insulin; measure expression of hypothalamic GLUT3and GLUT4geneand protein by real-time PCR and Western blot. The experimental data to mean±standard deviation express (X±s), using SPSS13.0for Windows for data processing,using analysis of variance, t test to compare differences between both, P<0.05for thedifference was significant.Results:1. The screening T2DM rats model with high fat diet for4weeks combined with lowdose STZ induced, compared with the blank group, model group and acupuncture groupFPG increased significantly (P<0.01), which suggests the model is successful. After2weeks treatment, FPG, FINS of model group and acupuncture group decreasedsignificantly (P<0.01,P<0.05).2. Model group rats of Quadriceps muscle muscle glycogen were significantlydecreased compared with those of blank group (P<0.01); the rats of acupuncture groupincreased compared with the model group, but no statistical significance.3.Expression of GLUT3mRNA and GLUT3protein in hypothalamus of rats weresignificantly decreased compared with those of blank group (P<0.01);Expression ofGLUT3mRNA and GLUT3protein in acupuncture group were increased significantlycompared with the model group rats (P<0.05,P<0.01).Expression of GLUT4mRNA andGLUT4protein in hypothalamus of rats were significantly decreased compared withthose of blank group (P<0.01); the expression of GLUT4mRNA and GLUT4protein inacupuncture group were increased significantly compared with in the model group rats(P<0.05,P<0.01).Conclusion:1.Acupuncture can significantly reduce the T2DM rat fasting blood glucose、fasting insulin. Acupuncture can correct the T2DM rat endocrine disorders, improve insulinresistance, which is an effective method for the treatment of type2diabetes mellitus.2. Acupuncture can effectively increase the expression of GLUT3,GLUT4mRNAandGLUT3, GLUT4protein expression of the T2DM rats hypothalamus.Acupuncturecan improve the central insulin resistance in transduction of postreceptor defects,increase the content of hypothalamus glucose transporter, enhance hypothalamicglucose transport capacity.3. The T2DM effective mechanism of acupuncture treatment might be throughmulti-channel, multi-level to improve insulin resistance.
