Objective:To explore that whether or not glycyrrhizic acid can antagonise the neurotoxicity caused by aconitine and the mechanism that glycyrrhizic acid antagonise the neurotoxicity.Methods:Neuron cells were prepared from neonatal SD rats and cultured in vitro. The MTT assay was used to determine how mixture of glycyrrhizic acid and aconitine in different proportions (1:1、1:2、1:0.5) infect the survival of nerve cell in primary culture.The turbidimetric method, enyme method and ELISA were used to study the effect of the compatibility of glycyrrhizic acid and aconitine on membrance, internal environment, energy metabolism and2kinds of neurotransmitters. And to study if glycyrrhizic acid can antagonise the neurotoxicity of aconitine or not.Results:After administration by the0.2%aconitine for30min, the survival rate of nerve cell were decreased, and the activity of Na+-K+ATPase and the contents of [K+] and [Ca2+] in the cells were increased. The contents of LDH in culture medium, the activity of ACP and MDA in cells and the release of GAB A were all increased, while the activity of COX and the release of ACh were decreased. The activity of Ca2+-Mg2+ATPase and the contents of [Na+] and [Mg2+] in the cells were decreased. The mixture of glycyrrhizic acid and aconitine with different proportions can alleviate the changes caused by aconitine.Conclusions:After administration on neuron cells for30min, the compatibility of the glycyrrhizic acid and aconitine in different proportions can antagonise the neurotoxicity caused by aconitine. And maybe the glycyrrhizic acid can protect neuron cells. The mechanism were to improve the energy metabolization, protect the cell membrance, regulated the release of neurotransmitters and inhibit the disturbance of the internal enviroment, especially the overload of intracellular. In addtion, the best compatibility proportion is1:2.
