Evaluation Of Diagnostic Value Of Inter-α Inhibitor H1, Serpin Peptidase Inhibitor Clade F Member2, Transthyretin In Hepatic Fibrosis And Which Affected By Fufanghanfangji

ObjectiveTo explore the expression of inter-α inhibitor H1（ITIH1）,serpinpeptidase inhibitor clade F member2（SERPINF2）,transthyretin（TTR）in serum and liver tissues on rats with liver fibrosis induced by CCl4and to explore the diagnostic value of ITIH1,SERPINF2,TTR in liverfibrosis;And to study the effect of Fufanghanfangji on the expressionof ITIH1,SERPINF2,TTR and to define the therapeutic effect ofFufanghanfangji’s anti-liver fibrosis.MethodsThirty male Fisher344rats were randomly divided into normalcontrol group, model group, Fufanghanfangji low-dose group,medium-dose group and high-dose group.The animal models of liverfibrosis were induced by subcutaneously injecting with40%CCl4（2.5ml/kg）,twice a week for18weeks.Except group A,all the othergroups were injected with40%CCl4（2.5ml/kg）,twice a week for18weeks. Group A was only injected with physiological saline（2.5ml/kg）,The Fufanghanfangji compound intervened groups weregiven supplemental Fufanghanfangji （143mg/kg,286mg/kg,572mg/kg）by gastric gavage,once every other day,18weeks at the sametime when establishment of models began,the model group was onlygivenphysiological saline by gastric gavage,once every otherday,lasting18weeks.The expression of ITIH1,SERPINF2,TTR were detected in serum and liver tissues with ELISA method andImmunohistochemical stainingand,and the mRNA expression in thetissues were detected by RT-PCR.Liver fibrosis were evaluated onthe liver tissue sections which were performed with HE,Gordon-Sweet and Masson staining.ResultsCompared with normal control group， the expression of ITIH1,SERPINF2,TTR in liver tissues and serum was depressed obviously inmodel group （P<0.01）. Compared with model group, ITIH1, SERPINF2,TTR were significantly higher in supplemental Fufanghanfangjigroup（P<0.05orP<0.01）. The degree of liver fibrosis in Fufanghanfangji intervened group was obviously lowered.ConclusionsITIH1,SERPINF2,TTR can reflect degree of hepatic fibrosis,theymay paly an important role in the diagnosis and monitoring of liverfibrosis;Fufanghanfangji has the positively preventive effects on theexperimental liver fibrosis induced by CCl₄in rats and can significantlyincrease the expression of ITIH1,SERPINF2,TTR.
