Purpose:Up to now,Chronic Obstructive Lung Disease(COPD) has became one of the most seriously diseases that harmful to the broad masses of the people,especially to the health of elderly people.It has been demonstrated by sorts of studies that COPD was a complex disease affected by both the genes and environmental factors.Expect the environmental factors,smoking is an important risk factor which was recognized by scientists.However,only a few of smokers suffers COPD.In a study on early-onset COPD family in Boston,It has been manifest that the long arm of chromosome 2 was strongly associated with COPD by Genomewide linkage analysis according to lung function test.DeMeo and his colleagues discovered SERPINE2(serine protease inhibitor-2 or plasminogen activator inhibitor1) lied on the 2q was a predisposing gene of COPD.Our purpose was to investigate the dependability of SERPINE2 gene polymorphism in serum and Chronic Obstructive Pulmonary Disease(COPD) phenotypic in order to identify the pathogenesy of COPD.Materials and Methods:We adopted case control study.We collected 100 Han people inpatient or outpatient in Shandong university Qilu hospital form October,2007 to June,2008 as the experiment group,all of them are consistent with the diagnoses and treatment guideline of Chronic Obstructive Pulmonary Disease published in 2007.The forced expiratory volume in one second account to prediction<70%after patients inhalation 200ug Salbutamol..We chose 100 cases taken health examination and were totally healthy as control group.They all had normal results of chest X-ray, and their FEV1/FVC>70%,FEV1 account to the prediction 80%.We extracted the genome DNA,designed the common TaqMan probe and primer using Primer Express 2.0 software.We detected the single nucleotide polymorphism(SNP) of two groups through allele specific probe, to determine the genetype of single sample.Results:the mutation of rs6734100 site is not relevance to COPD.The mutation frequency of cases group and control group is 2.5%and 0.0%, there are no statistically significant between the two group byχ2 analysis(p<0.05).We divided patients intoⅠ,Ⅱ,Ⅲ,Ⅳgrade according to lung function.(view the guideline for COPD 2007) and there are no statistically significant between the four grade byχ2 analysis(p<0.05).The analysis of coefficient correlation of the Smoking index,SERPINE2 rs6734100 polymorphism and COPD,the coefficient correlationγbetween smoking index and COPD is 0.208(P=0.112),the coefficient correlationγbetween SERPINE2 rs6734100 polymorphism and COPD is 0.056(P=0.787).Both P value>0.05,there are not statically significant.Conclusion:SERPINE2 is not a predisposing gene of COPD Han people. There has demonstrated that SERPINE2 mutation site is relevance to COPD by linkage analysis of the mice and human 2q domain gene information. There is not yet SERPINE2 gene polymorphism and incidence of COPD-related research at present in our country.This study compared similar experiment abroad by detecting the SERPINE2 rs6734100 polymorphism,demonstrated that the mutation of SERPINE2 rs6734100 hasn't obviously relevance to COPD.
