| ObjectiveTo observe the effects of six antipsychotic drugs, haloperidol, clozapine, olanzapine, quetiapine, risperidone, and ziprasidone on the differentiation of3T3-L1preadipocytes and on the level of adiponectin protein expression in mature fat cells.Methods1.3T3-L1preadipocytes were in vitro recovered, cultured, and passaged with standard processes, and were routinely induced to differentiation by the hormonal cocktail. Oil red staining was used to identify the fully differentiated adipocytes.2.3T3-L1preadipocytes were induced to differentiation in treated groups containing haloperidol, clozapine, olanzapine, quetiapine, risperidone, and ziprasidone with different concentrations (0.1μmol/L,1μmol/L,10μmol/L,100μmol/L), an set control groups and vehicle (DMSO) control groups. Then, at the8th day, after oil red O staining, cells were observed under microscope and taken pictures to record the resault. 3. Mature3T3-L1fat cells were intervention for24hours and48hours respectively in treated groups containing drugs with different concentrations (0.1μmol/L,1μmol/L,10μmol/L,100μmol/L), and set control groups and vehicle (DMSO) control groups. Then detected the level of adiponectin protein expression in cells by Western Blot.Resault1.3T3-L1preadipocytes were induced into mature adipocytes by hormonal cocktail successfully.2. By Oil red O staining we found that the differentiation rate of haloperidol, clozapine, olanzapine, quetiapine, ziprasidone with lower concentration intervention cells was significantly higher than that of vehicle control groups (P<0.05), but with the concentration of drugs increasing, the number of lipid droplets decreasing in cells; The differentiation rate of the aforementioned five drugs with higher concentration intervention cells are significantly lower than that of vehicle control groups, and with the concentration increasing, the number of lipid droplets decreasing. But the differentiation rate of risperidone with any concentration intervention cells was significantly lower than that of vehicle control group (P<0.05).3. Western Blot showed that level of adiponectin protein expression in cells that all the six drugs intervention with different concentrations had no statistically significant difference (P>0.05). Conclution1. Antipsychotic drugs haloperidol, clozapine, olanzapine, quetiapine, ziprasidone with lower concentrations could promote the differentiation of3T3-L1preadipocytes, but they could inhibit it with the higher concentrations; But risperidone could inhibit the differentiation with any experimental concentrations.2. The six antipsychotic drugs may not directly impact on level of adiponectin protein expression in mature fat cells. |
