Objective:To observe the effect of Leptin(LP) on the expression of C-reactive protein(CRP) and Plasminogen activator inhibitor-1(PAI-1) in the human coronary artery endothelial cells(HCAECs). To investigate the relationship between LP and coronary thrombosis.Methods:Experiments were performed using primary human coronary artery endothelial cells. All experiments were performed at3-5passages with70-80%confluency after overnight incubation in serum and growth factor free media, then divide them into six groups and make six holes in every group. The control group with implement was DMEM which didn’t use endothelial cell growth factor.while the left five groups ware operated with Leptin LP in different concentrations (Ong/ml、20ng/ml、50ng/ml、100ng/ml、200ng/ml).This period lasted48hours. In order to gather condition culture medium, enzyme linked immunosorbent assay(ELISA) was used to test the saturation of CRP and PAI-1。Western Blot(WB) was used to test the protein expression of CRP and PAI-1in the cells.Results:(1) After comparing the levels of CRP in the six cell culture mediums, we found that the levels of CRP in different concentrations of Leptin(50-200ng/ml) were all higher than that in the control group, whose inductive effected grew stronger with the concentration of Leptin increasing. The difference was meaningful in statistics(P=0.000,0.000,0.000,<0.05).(2) After comparing the levels of PAI-1in the six cell culture mediums, we found that the levels of PAI-1in different concentrations of Leptin(50-200ng/ml) were all higher than that in the control group, whose inductive effected grew stronger with the concentration of Leptin increasing. The difference was meaningful in statistics (P=0.000,0.000,0.000,<0.05).(3) After comparing the comparative expressions of CRP in the six cell culture mediums, we found that their expressions in different concentrations of Leptin(50-200 ng/ml) were all higher than that in the control group, whose inductive effected grew stronger with the concentration of Leptin increasing. The difference was meaningful in statistics (P=0.000,0.000,0.000,<0.05).(4) After comparing the comparative expressions of PAI-1in the six cell culture mediums, we found that the expressions in different concentrations of Leptin(50-200ng/ml) were all higher than that in the control group, whose inductive effected grew stronger with the concentration of Leptin increasing. The difference was meaningful in statistics (P=0.000,0.000,0.000,<0.05)Conclusions:Leptin could show dose dependent increase in the inducement of the expression of CRP and PAI-1protein in human coronary artery endothelial cells. This indicated that leptin had a proinflammatory effect and leptin could Promote thrombosis. This was as an important cause of an independent cardiovascular risk factor, and prompt leptin was a potential drug targets for intervention in cardiovascular disease.7Figures,4Tables,44References.
