Objective:To investigate the expression of the transcription factor Sox2and the cyclin-dependent kinase inhibitor p27Kip1in human gliomas. Analysis the expression of Sox2and p27Kip1and its relation with the malignancy of the tumor, explore their roles in the Origin of gliomas and glioma stem cells.Methods:The expression of Sox2and p27Kip1were detected by immunohistochemical method (SP) in65human glioma samples and12normal brain tissues which all came from the frontal lobe. Carry on comprehensive analysis with the clinical pathologic parameters.Results:(1)The expression level of Sox2in normal brain tissue was2.33%±1.61,not reach the positive standard.We found Sox2positive cells in all65human glioma samples,only4did not meet the positive criteria. The expression level of Sox2in glioma samples from grade I to IV were4.30%±3.05,16.22%±7.16,18.80%±7.08and54.15%±13.82. The analysis of variance indicate the disparity between each group has statistic meaning (F=88.114,P<0.01).Through SNK-q test we can found that the expression level of Sox2in High-grade gliomas (grade IV) was significantly higher than the low-grade gliomas (grade I) and normal brain tissue (P<0.01),so the expression level of Sox2maybe related to the malignancy of the tumor.But there were no difference between the expression of Sox2in normal brain tissue and grade I glioma samples as well as in grade II and III glioma samples (P>0.05)(2) The nuclear expression level of p27Kip1in human glioma samples from grade I to IV were62.60%±9.65,50.59%±10.52,35.20%±11.12and12.0%±4.32which lower than73.42%±13.77in the normal brain tissue. The analysis of variance indicate the disparity between each group has statistic meaning (F=68.0,P<0.01).Through SNK-q test we can found that the nuclear expression of p27Kip1decreased with the increase of grades of gliomas, the disparity has statistic meaning (P<0.01).(3) Handling with linear correlation, the expression level of Sox2was negatively correlated with p27Kip1in human glioma samples (r=-0.75, P<0.01)Conclusions:(1) Sox2was overexpressed in human glioma samples whereas expression in normal brain tissue was almost negligible. And the expression level of Sox2maybe related to the malignancy of the tumor;(2) The nuclear expression level of p27Kip1in human glioma samples related to the malignancy of the tumor, decreased with the increase of grades of gliomas;(3) The expression level of Sox2was negatively correlated with p27Kip1in human glioma samples.They may be involved in the cellular Origin of gliomas.
