Objective:①Assessment the curative effect for adult neural stem cells transplanted into SD rat with cerebral hemorrhage.②observe the differentiation direction of the ANSC in the rat’s brain.Methods:③Get the adult SD rat’s hippocampal dentate gyrus, preparate the single cell suspension by mechanical percussion and trypsinization;Collect the ANSCs by the way of density gradient centrifugation and special culture medium.②Get twenty rats with intracerebral hemorrhage by injecting the autoblood into the striatum, than select ten rats with the same score,randomly divide it into two groups including experimental group and control group;Twenty-four hours later,inject the singed ANSCs into the experiment group and saline into the control group.③After transplantation,observe the recovery of the rats’s neural function by the way of rotation and placement of limbs every week for seven times,than detect the differentiation direction of the ANSCs in the brain by immunohistomical seven weeks later.Results:④The ANSCs grow spheraityly,it begin to differentiate and the cells in the core begin to die when the spherait diameter close to160um,it can keep high proliferation activity when passaged to the third generation.②After injecting the autoblood in to the striaum of the rat,it can performance the symptoms of intracerebral hemorrhage.③Seven weeks after transplantation, the ANSCs remarkbbly promote functional recovery of the rat,some of the ANSCs are BRDU/GFAP+and BRDU/NEUN+.Conclusion:①We can separate the ANSCs in the rat’s hippocampal dentate gyrus.②We can get stable intracerebral hemorrhage stroke models by injecting the autoblood into the striatum.③The ANSCs can remarkably improve the neural function of the rat,and in the seventh week, it can differentiate into cells which express special markers of neurons and gliacyts.With thirteeen figures,two tables and forty-eight references.
