Objective:To explore the effects of electro-acupuncturecombined with BMSCs transplantation to the lesion surroundingmicroenvironment in the rats with intracerebral hemorrhage, to explorethe mechanism of neuron repairMethods:1. When the cells that BMSCswere recovered adherent up to80%, replaced with the pre-inductionmedium and cultured for24hours, and then replaced by serum-freeinduction medium induced6h. The cells were collected for using.2.Theanimal model of intracerebral hemorrhage was established by injectingCollagenase and heparin into the Candate-Putamen in brain of ratsthrough the brain Stereotaxic apparatus.3The neurological deficits wereestimated by NSS scores after surgery until the rats awake at2h,24h,48htime point.The rats which score was more than9points was divided intothe experiment. The experimental animals were divided into three groups:normal saline group (control group), BMSCs transplantating group(BMSCs group),BMSCs transplanting combined withelectro-acupuncture treatment group (Ea BMSCs group).3.Through brainStereotaxic apparatus,20ul0.9%normal saline and BMSCs suspensionwere transplanted into ICH lesions in3rdday after operation. According to different feeding times,each group was divided into5subgroups(1d,3d,5d,7d,14d). Electro-acupuncture stimulated the DaZhui and BaiHuiafter2h of transplantation, and the electro-acupuncture treatment once aday until the rats death.5.Recording the NSS scores of each group untillthe rats was killed for material.6.observing the protein expression ofNGF,BDNF,-tubulin Ⅲ and GAPDH by western blot.Results: after thesuccess of rat ICH model, recording the NSS score of control group,BMSCs group and Ea BMSCs group at the same time point before cellstransplanted. There is no significant difference between groups.5daysafter transplantation, there is significant differences between the BMSCsgroup and the control group in NSS scores(P<0.05). Ea BMSCs groupcompared to control group (except1d) have significant differences(P<0.05). and there is more obviously to improve the symptoms ofneurological deficit.2.the NGF protein expression in Ea BMSCs groupand BMSCs group was significantly increased. After7and14days,BMSCs group increased significantly compared with control group (P<.0.05). Ea BMSCs group compared with BMSCs group and controlgroup (except1d) have more significant difference (P <0.05). the NGFprotein expression increased more significantly in Ea BMSCs group ofrats with intracerebral hemorrhage around the lesion.3. BDNF proteinexpression was significantly increased in Ea BMSCs group and BMSCsgroup at5,7and14days after transplantation. There is significant differences compared with control group (P <.0.05). Ea BMSCs groupcompared to BMSCs group have more increased protein expression. wasstatistically significant (P <0.05). after5,7and14days aftertransplantation.Ea BMSCs group compared with BMSCs group andcontrol group the BDNF protein expression increased more significantlyin the rats with cerebral hemorrhage around the lesion.4.At the same timepoint, the Ea BMSCs group and BMSCs group compared to control groupthe β-tubulin Ⅲ protein expression was significantly increased; after5,7and14days transplantation, the protein expression in BMSCs groupincrease significantly than the control group, there are statisticallysignificant (P <0.05);, Ea BMSCs group (except1d) compared withBMSCs group increased significantly(P <0.05). electro-acupuncturecombined with BMSCs transplantation can up-regulated the NGF andBDNFprotein expression in the lesion of ICH, improve the surroundingmicroenvironment and promote neuronal repair.Conclusion:1,Transplanted BMSCs can recover the hemorrhage of rat brain byup-regulated the NGF and BDNF protein expression around the braintissue to protect neurons.NGF and BDNF protein expression in theduration of certain repetitive.2.Electro-acupuncture combined withBMSCs transplantation can improve the neurological deficit symptoms ofintracerebral hemorrhage in rats, and to promote its motor and sensoryfunction recovery than a simple transplanted BMSCs3. Electro-acupuncture combined with BMSCs transplantation to improvethe lesion surrounding microenvironment in the rats with intracerebralhemorrhage and promote endogenous and exogenous neuronal survival4.The ability of neurological function recovery in rats which sufferd ICH isstrong..
