Objective:To establish model of osteoarthritis rats, through detecting the expression of Wnt7a and β-catenin in model joint to discuss the effect that Wnt7a and β-catenin to the joint in the occurrence and development of osteoarthritis and the possible presence of machanism action.Methods:To divide40healthy SD rats into2groups with the same numbers by randomly, one group is named experiment, the other is control group. The rats in experimental group injected UPA to the rats’ knee joint cavity to built osteoarthritis model, while the rats in control group injected saline in the same place and with the same capacity. After eight weeks, to implement anaesthesia on rats with lethal dose and then collecting specimens under aseptic condition. Paraffin embedding after fixing the collected specimens, immunohistochemical staining after sectioning. Measuring the mean gray value with IPP image analysis software and analysing groups’data with statistical safeware SPSS17.0Results:1. The mean gray value of Wnt7a in cartilage is normal,140.20±8.85for control group and123.10±14.80for experimental group.There is obvious difference (P<0.05) between these two data in statistics. The mean gray value of β-catenin in cartilage is normal,127.00±9.18for control group and110.27±12.50for experimental group, there is obvious difference (P<0.05) between these two data in statistics. The mean gray value’s expression of Wnt7a and β-catenin in cartilage is ralative (r=0.622, P<0.01).2. The mean gray value of Wnt7a in synovial is normal,159.10±8.65for control group and144.06±6.81for experimental group. There is obvious difference (P<0.05) between these two data in statistics. The mean gray value of β-catenin in synovial is normal,145.80±3.73for control group and132.76±12.28for experimental group, there is obvious difference (P<0.05) between these two data in statistics. The mean gray value’s expression of Wnt7a and β-catenin in synovial is ralative (r=0.879, P<0.01)3. The mean gray value of Wnt7a in subchondral bone is normal,123.30±10.15for control group and113.13±13.10for experimental group. There is obvious difference (P<0.05) between these two data in statistics. The mean gray value of β-catenin in subchondral bone is normal,137.70±9.41for control group and113.16±8.42for experimental group, there is obvious difference (P<0.05) between these two data in statistics. The mean gray value’s expression of Wnt7a and β-catenin in subchondral bone is positively ralative (r=0.672, P<0.01) Conclusion:1.In the rats’osteoarthritis, the expression of Wnt7a and P-catenin extremely increased in cartilage, synovial and subchondral bone.2.In the rats’osteoarthritis, the expression of Wnt7a and β-catenin in cartilage, synovial and subchondral bone is positively relative which prompted Wnt7a may play role through regulating the expression of β-catenin to involve the occurrence and development of osteoarthritis.
