A Study About AP-1Interacting With NF-κB In Human Myometrium In Labor Situation Of Term Pregnancy

Objective:To verify the interaction between Nuclear factor kappa B and Activator protein1in human myometrium in labor situation by application of immunoprecipitation and Western blotting.Methods:The expression of NF-κB and AP-1were detected by Western blotting in labor group, non-labor group and non-pregnant group. Then immunoprecipitation separates and purifies the protein interacting with NF-κB in the three groups. At the same time, immunoprecipitation separates and purifies the protein interacting with IgG in the labor group. The objective is to enrich NF-κB and other transcription factors which interacts wit NF-κB.Then immunoblotting verifies AP-1interacting with NF-κB by anti-NF-KBp65antibody.Then immunoprecipitation separates and purifies the protein interacting with AP-1in the three groups. At the same time, immunoprecipitation separates and purifies the protein interacting with IgG in the labor group. The objective is to enrich AP-1and other transcription factors which interacts wit AP-1.Then immunoblotting verifies AP-1interacting with NF-κB by anti-c-Jun antibody.Results:1. The results of Western blotting show that nuclear proteins were extracted successfully. AP-1and NF-κB were contained in the total protein liquid of these tree groups.2. Immunoblotting verifies that AP-1interacts with NF-κB in the labor group and non-labor group from the protein complexes immunoprecipitated by anti-NF-κBp65antibody,but,the interaction of labor group is dramatically stronger than non-labor group. We have not seen bands in non-pregnancy group and IgG of labor group.3. Immunoblotting verifies that AP-1interacts with NF-κB in the labor group and non-labor group from the protein complexes immunoprecipitated by anti-c-Jun antibody.But,the interaction of labor group is dramatically stronger than non-labor group.We have not seen bands in non-pregnancy group and IgG of labor group.Conclusion:AP-1interacts with NF-κB in human myometrium around labor situation,and,the interaction is dramatically stronger in labor situation.
