Background:Colorectal Cancer (colorectal cancer, CRC) is one of commonmalignant tumors of the gastrointestinal carcinoma, and seriously threatening humanhealth. In China, the incidence and fatality rate have increased year by year, thecolorectal cancer is the fifth leading cause of death in malignant tumors, behind lungcancer, liver cancer, gastric cancer and esophageal cancer. As the most malignanttumors, the development and progression of colorectal cancer is a complex process thatmulti-factors and multi-stages involved in. With the rapid development of molecularbiology in recent years, a number of protein molecular biomarkers become the hot spotsin oncology research which applied to the treatment and prognosis of malignant tumors.The basic research of HER-2/neu gene has become a hot spot in this field. TheTrastuzumab which target therapy of HER-2/neu gene has been widely used in thetreatment of breast cancer and gastric cancer. However, for treatment of colon cancerhas not yet clear. To detect HER-2/neu gene expression in colon cancer tissues andparacancerous tissues, and analyze the relationship between clinical features andpathological types,The study is used through the immune histochemical method(IHC).And provide a reference for clinical treatment and assessment of prognosis.Objective: To test and discuss the expression of HER-2/neu gene in colon cancertissue and paracancerous tissues,investigate the pathological type, clinicalcharacteristics and relationship.Materials:To collect the clinical data and pathological paraffin blocks of January1,2008to June30,2012Second Affiliated Hospital of Dalian Medical University223routine colon cancer radical surgery patients.Methods: The immunohistochemical method is used to detect the370casesexpression of HER-2/neu gene in colon cancer. And χ2is adopted to test and analysisthe expression of HER-2/neu gene in colon cancer tissue, and the relationship between pathological type and clinical characteristics. To analysis by SPSS17.0software, WhenP<0.05, it means that the discrepancy makes sense in statistics.Results:1.HER-2/neu expressed as respectively by74cases and0cases in223cases ofcolon cancer and paracancerous tissues, the positive expression rate were33.2﹪and noexpression, adenocarcinoma and adjacent tissues of cancer group had significantdifference (P<0.05).2.In223cases of colon cancer tissues, papillary or tubular adenocarcinoma is182cases, the positive expression is60cases, positive expression rate is33﹪, mucousadenocarcinoma is41cases,the positive expression is14cases, and positive expressionrate is34.1﹪,The expression of HER-2/neu in different histological types of coloncancer had no statistical difference (P>0.05).3. The expression of HER-2/neu is not correlated with gender, age, tumormaximum diameter, gross classification, degree of tumor differentiation, depth ofinfiltration, it had no statistical difference (P>0.05). And the expression of HER-2/neuis correlated with metastasis of lymph node and the Dukes staging, it had statisticaldifference (P<0.05).4. The expression of HER-2/neu is positively correlated with the metastasis oflymph node and Dukes staging, The coefficient correlation of Spearman are respectively0.320and0.320. With the increasing number of lymph node metastasis, positiveexpression rate increases. The positive expression rate of Dukes staging in C and Dperiod is significantly higher than that of A and B period of the patients.5.The expression of HER-2/neu in74cases of the positive colon cancerpatients,60-74age group accounted for59.4﹪(44/74).The expression of HER-2/neu incolon cancer patients of the60-74age group is higher than the other groups. Familialadenomatous polyposis group accounted for97.3%(72/74), The expression ofHER-2/neu is higher than familial adenomatous polyposis group.Conclusion:1. The expression rate and expression level of HER-2/neu in colon cancer tissueare significantly higher than in normal tissue adjacent to carcinoma.2. The expression of HER-2/neu is not correlated with gender, age, differenthistological types, tumor maximum diameter, gross classification, degree of tumordifferentiation and the depth of infiltration. And the expression of HER-2/neu iscorrelated with metastasis of lymph node and the Dukes staging. 3. The expression of HER-2/neu is positively correlated with the metastasis oflymph node and Dukes staging. With the increasing number of lymph node metastasis,positive expression rate increases. The positive expression rate of Dukes staging in Cand D period is significantly higher than that of A and B period of the patients.4.The positive expression of HER-2/neu in colon cancer patients,60-74age groupand no familial adenomatous polyposis group are higher than other groups.
