Objective: Gastric cancer ranks in the second among malignant tumors in China.Currently, surgical operation is still considered as the preferred treatment choice ofgastric carcinoma. However, owing to the lack of specialized cancer filtration index andinconspicuous symptoms for early patients, most of the patients missed their optimalopportunity of surgery when they were clearly diagnosed. Even if excision is applicable,the postoperative local recurrence rate is up to80%after tumor resection within5years.Therefore, chemotherapy is considered as the conventional treatment, especially for thesufferers whose tumor is metastatic or can’t be resected. But meanwhile, as a result oftumor drug resistance, immunologic suppression and toxicity and side effects ofchemotherapy, the majority patients with weak constitution are failed to process theeffective chemotherapy periodically.Based on the recent investigation, traditional Chinese medicine has its ownadvantages in the treatment of gastric cancer. It is very efficacious because of its mildeffects, particularly to the elderly, the frail and the patients in advanced gastric cancerwho are not suitable for operation or chemotherapy. So the anti-tumor research incombined therapeutics of traditional Chinese and western medicine has become one ofthe research hotspots.The traditional Chinese medicine used in this paper (Jiayang decoction) isextracted on the page660of "Secret Recipes of China" which is compiled by ZhouHongfan. The main ingredient of Jiayang decoction is Scutellaria barbata andOldenlandia diffusa, which plays a significant role in enhancing immune function, inhibiting proliferation of tumor cell, promoting apoptosis of tumor cells and restrainingtumor neovascularization, etc.In this study, we intend to combine Jiayang decoction with Oxaliplatin and apply itto gastric cancer cells SGC-7901in vitro, and then investigate its potential mechanismby observing the inhibition effect on the growth of cell SGC-7901by comparing theresults in different groups of Jiayang decoction, Oxaliplatin and their combination, so asto provide experimental evidence for further clinical application.Methods: There are4test groups of Jiayang decoction group, Oxaliplatin group,the combination group and control group. Plan to use following methods to detect thegrowth condition of gastric cancer cell SGC-7901in vitro after they were affected bydifferent concentration of Jiayang decoction and Oxaliplatin alone and by thecombination of the two drugs:(1) adopt MTT colorimetric assay to test and analyzeinhibition of proliferation;(2) use inverted microscope to observe the morphologicalchanges of cells. All measurement data are expressed as mean values±standarddeviation (SD). Variance between the groups was analyzed by performing T-test. Thestatistical significance for all P values was defined as P<0.05.Results:(1) Jiayang decoction could inhibit the proliferation of cell SGC-7901,and the suppression depends on the action time and drug concentration.(2) It showssynergistic effects after48hours when combine Jiayang decoction of50μl/ml withOxaliplatin of1μg/ml, and the inhibition rate of combined group was observably higherthan that of decoction group and Oxaliplatin group.(3) Observed cells under invertedmicroscope, their configuration have changed after the action of the drug. The volumebecomes smaller and the cell gap becomes looser gradually.Conclusions:(1) Jiayang decoction has the effect on proliferation suppression togastric carcinoma cell line SGC-7901, and the inhibition depends on the action time anddrug dose.(2) It strengthened the inhibition when Jiayang decoction combined withOxaliplatin, and their combination has a synergistic effect on the growth repression ofgastric cancer cells.
