| Objective:To investigate the inhibited effect and anti-tumor mechanism of combined Hedyotic diffusa and Scutellaria barbata superfine powder on H22 hepatoma-bearing mice. Methods:By building a mouse model of transplanted hepatoma H22, adopting orthogonal experimental L9 (34) design table and using tumor inhibition rate,the most effective compatibilities group of Hedyotic diffusa and Scutellaria barbata superfine powder is selected. Observe the effects of Hedyotic diffusa superfine powder group,Scutellaria barbata superfine powder group and compatibility groups (A2B3,A3B2,A3B3) on tumor inhibition rate, thymus and spleen index of H22 tumor-bearing mice; observate the morphological changes and PCNA,Bcl-2,Bax expression of tumor cells by pathomorphology and immunohistochemistry methods; observate the effects on cell cycle, apoptosis and proliferation of tumor cell by flow cytometry. And compare the differents between compatibility groups and Hedyotic diffusa or Scutellaria barbata group.Results:By orthogonal experimental design, find out that group A3B3 of Hedyotic diffusa and Scutellaria barbata superfine powder exhibits the highest tumor inhibition rate. From nine groups, choose out group A2B3,A3B2,A3B3 which have over 40 percent of tumor inhibition rate to investigate the anti-tumor mechanism.Compared with model group,the WBC count of compatibility group A3B3 is significantly increased,also than used Hedyotic diffusa singly; the thymus and spleen index of group A2B3,A3B3 are significantly improved. The tumor cells of group A2B3,A3B2,A3B3 have an increase in the cytoplasmic density aggregated lumps in chromatin, and can find apoptotic bodies. Compared with model group, the Bax positive ratios of medicine groups are increased, the Bcl-2 are decreased significantly; the PCNA positive ratios of group A2B3,A3B2,A3B3 are decreased significantly; and the effect of group A3B3 is better than using Hedyotic diffusa or Scutellaria barbata singly on the three ratios. Compared with model group,in group A3B3,the number of G0/G1 phase cells increase significantly, the number of S phase cells decrease significantly, and the tumor cells are blocked to GO→S phase;the proliferation index obviously decreases; the percentage of apoptotic cells obviously increases and better than using Hedyotic diffusa or Scutellaria barbata singly. Conclusion:By enhancing immune function, effecting cell cycle, decreasing proliferation and inducing apoptosis of tumor cells,Hedyotic diffusa and Scutellaria barbata superfine powder compatibility give play to inhibiting tumor in H22 tumor-bearing mice,and better than using Hedyotic diffusa or Scutellaria barbata singly. |
