Aconiti Radix and Aconiti Kusnezoffii Radix were a toxic herbs and used widely, both had similar effect, TCM holded that they had the ability of expelling wind, removing damness, warming channels and relieving pain. Modern pharmacological studies had shown that Aconiti Radix and Aconiti Kusnezoffii Radix had the effect of anti-inflammatory, analgesic, cardiov-ascular system, immune system, anti-tumor, lowering blood sugar and so on. But these kinds of Chinese medicine was highly toxic, so it must be processed before taking, and there was many kinds of methods for processing of Aconiti Radix and Aconiti Kusnezoffii Radix, leading to variable quality in market, affecting the clinical safety and efficacy of medication. This experiment conducted the following aspects for Aconiti Radix and Aconiti Kusnezoffii Radix: 1 Determination the content of index components of Aconiti Radix, Aconiti Kusnezoffii Radix and their processed productsExperiments established the determination method of the index com-ponent of Aconiti Radix, Aconiti Kusnezoffii Radix and their processed products, using the method of HPLC to determine the content of benzoylmes-aconine, benzoylaconine, mesaconitine, hypaconitine, aconitine of differ-ent origin Aconiti Radix, Aconiti Kusnezoffii Radix and their processed products, and determine the content of total alkaloids and esterifiable alkaloids by UV. The results showed that the content of six kinds of alkaloids, total alkaloids, esterifiable alkaloids were significantly different of Aconiti Radix, Aconiti Kusnezoffii Radix and their processed products, so we should establish a scientific and reasonable method for quality control to ensure their quality. 2 The study of processing technology of Aconiti Radix and Aconiti Kusnezoffii RadixExperiments determined the content of six kinds of alkaloids of different processed products of Aconiti Radix and Aconiti Kusnezoffii Radix by HPLC, and using the method of UV to determine the content of total alkaloids. The content of six kinds of alkaloids and total alkaloids were determined as the investigative index, to study the processing technology of softening way, high-pressure steaming, high temperature baking, microw-ave, and compare four kinds of processing methods with the methods of the edition 2010 of "China Pharmacopoeia" about processed products of Aconiti Radix and Aconiti Kusnezoffii Radix, finally, we synthetically evaluated their processing technology. The results suggested that the best high-pressure steaming processe of Aconiti Radix and Aconiti Kusnezoffii Radix was that:after wetting with waters, processing 150 min under high pressure at 1.5 kg·cm-2; The best high-temperature baking processe of Aconiti Radix and Aconiti Kusnezoffii Radix was that:after treatment with the method of cutting after moistening, baked at 110℃temperature 8 h and 10 h, respect-tively; The best microwave processing technology of Aconiti Radix and Aconiti Kusnezoffii Radix was that:after treatment with the method of cutting after moistening, and cooked 18~20 min and 25 min under microwave heating with 60%, respectively; Comparing to the method of the edition 2010 of "China Pharmacopoeia" about processed products of Aconiti Radix and Aconiti Kusnezoffii Radix, all above three kinds of processing technology had some advantages, there were certain practical nature, can be used as new technology for Aconiti Radix and Aconiti Kusnezoffii Radix. 3 Study on processing principle for aconitum carmichaeliThe reference standard of benzoylmesaconine, benzoylaconine, benzoy-lhypaconine, mesaconitine, hypaconitine, aconitine were hydrolyzed und-er constant temperature, and using the method of HPLC determined the content of their hydrolysis products, to explore the law of its hydrolysis, and determined the content of main ingredients of Aconiti Radix and Aconiti Kusnezoffii Radix before and after processing, to explore the processing principle. The results showed that in certain conditions, diesterifiable alkaloids could into monoester alkaloids, there would produce an interme-diate product in hydrolysis process, we concluded that this intermediate product may be the type of pyraconitine. It was found that the hydrolysis process of aconitum alkaloids was not a simple process, and their hydrolysis law remains to be further studied. 4 Establishment of chromatographic fingerprint spectrum of Aconiti Radix, Aconiti Kusnezoffii Radix and their processed productsExperiments used the method of HPLC to establish the fingerprint of Aconiti Radix, Aconiti Kusnezoffii Radix and their processed products, identified the main peaks for the fingerprint and classified 3 areas of non-ester type, ester type, monoester type based on the retention time, it provide a basis for the species identification and quality control of Aconiti Radix, Aconiti Kusnezoffii Radix and their processed products, and provide a methodological basis for quality standards of them.This study established the determination methods of the index compo-nent of Aconiti Radix, Aconiti Kusnezoffii Radix and their processed products, according to reported literatures and the needs of industrial production, this experiment systematically studied the processing techno-logy of Aconiti Radix and Aconiti Kusnezoffii Radix and optimized a more scientific and rational processing technology for them. At the same time, we established the HPLC fingerprint for Aconiti Radix, Aconiti Kusnezoffii Radix and their processed products, it provide a basis for the species identification and quality control of Aconiti Radix, Aconiti Kusnezoffii Radix and their processed products.
