| Purpose:The purpose of this experiment is to investigate the effect of intracerebral hemorrhage lesions by using of traditional Chinese medicine treatment under the guidance of the blood clot removing and brain essence filling method of bFGF, to further study the impact of the endogenous neural stem cells by the treatment. This essay shall discuss whether the blood clot removing and brain essence filling method can increase positive expression of basic fibroblast growth factor and endogenous neural stem cells from ICH damaged surrounding brain tissue, make evidence for the efficacy of traditional Chinese medicine in hemorrhagic stroke by the modern laboratory testing methods, while the accumulation of the authentic experimental science information to further improvement and development of traditional Chinese medical science "Brains theory".Methods:Select clean and healthy, Wistar rats, weight between250~320g, male and female rats each for50%.randimize them into low dose group, Mid-dose group and the high dose group. Create low dose group, Mid-dose group and the high dose group of Chinese medicinal formulae for hemorrhagic stroke by the principle of random which received drug treatment. Create blank group, sham operation group, ICH model group for the control group. And divided them into three subgroups:Id,3d and7d group. Guarantee6rats in each group.Blank group without any treatment; sham group rat head use stereotactic positioning technology, using50μl saline injected into the brain and in the brains6.0mm and5.0mm at each needle retention30min and10min, then pull out the needle slowly. The remaining four group rats head use stereotactic positioning, taking blood from their own tail artery and then injected into the brain to create the ICH Rats. Target at the bleeding lesions to the rat brain injected into the arterial blood from the tail of50μl and in the brains6.0mm and5.0mm at each needle retention30min and10min, then pull out the needle slowly. Give the experimental animals normal feeding in the same environment. Take brain tissue after formalin fixation at each subgroup modeling point in time one day, three days and seven days. Optical microscopy analysis expression of bFGF and Nestin of the surrounding brain tissue in ICH Rats lesions.Results:(1) The Chinese medicinal formulae for hemorrhagic stroke which formulated by the blood clot removing and brain essence filling method can effectively reduce the hematoma area of the ICH Rats, which has better results than the ICH Rats group. It prompts that the Chinese medicinal formulae played a different degree of improvement for the ICH lesions environment. P<0.05;(2)The Chinese medicinal formulae for hemorrhagic stroke which formulated by the blood clot removing and brain essence filling method can icrease expression of bFGF.There are significant differences compared with the ICH Rats group, the high dose group and the Mid-dose group have obvious effect. p<0.01;(3)The Chinese medicinal formulae for hemorrhagic stroke which formulated by the blood clot removing and brain essence filling method can icrease expression of Nestin.There are significant difference compared with the ICH Rats group, the high dose group and the Mid-dose group have been effected obviously. p<0.01.Conclusion:There is the possibility of the Chinese medicinal formulae for hemorrhagic stroke which formulated by blood clot removing and brain essence filling method promoting the proliferation of endogenous neural stem cells by increasing basic fibroblast growth factorexpression, playing a role in the repair and regeneration of peripheral nerve tissue of theintracerebral hemorrhage lesions. |
