The Effect Of TRPV1on The Gastric Motility By Acupuncture

Objective:To observe the effect of (TRPV1) on the movement of mice gastric motility by acupuncture at acupoint " Zhongwan "Method:We performed experiments on wild type (WT) mice, knockout mice(KO) in which the TRPV1gene was disrupted using standard gene targeting techniques and wild type mice with Capsazipine i.p.(CPZ).Movement of gastric was recorded by fixing an balloon into sinus ventriculi, then observe the effect of two minutes electro-acupuncture (1mA,2mA,4mA) and hand-acupuncture at acupoint "Zhongwan".Result:l.In both wide type mice and TRPV1gene knockout mice, EA1mA at "Zhongwan" can remarkably decrease intragastric pressure(P<0.05); In CPZ, EA1mA at "Zhongwan" have little effect on intragastric pressure (P>0.05). Comparisons between groups were tested by One-Way ANOVA analysis and LSD test, there was no significant difference among groups2.In both wide type mice and TRPV1gene knockout mice, EA2mA at "Zhongwan" can remarkably decrease intragastric pressure(P<0.05); In CPZ mice, EA2mA at "Zhongwan" slightly decrease intragastric pressure (P>0.05). Comparisons between groups were tested by One-way ANOVA analysis and LSD test, there was no significant difference among groups (P>0.05).3.In both wide type mice and TRPV1gene knockout mice, EA4mA at "Zhongwan" can remarkably decrease intragastric pressure(P<0.05); In CPZ mice, EA4mA at "Zhongwan" slightly decrease intragastric pressure (P＞0.05), Comparisons between groups were tested by One-Way ANOVA analysis and LSD test, There was a significantly difference among groups (P<0.05). It showed that the intragastric pressure decrease is:WT group=KO group> CPZ group4.In both wide type mice and TRPV1gene knockout mice, hand acupuncture at "Zhongwan" can remarkably decrease intragastric pressure(P<0.05); In CPZ mice, hand acupuncture at "Zhongwan" slightly decrease intragastric pressure (P>0.05), Comparisons between groups were tested by One-Way ANOVA analysis and LSD test, There was a significantly difference among groups (P<0.05). It showed that the mount of intragastric pressure decrease is:WT group>KO group> CPZ group.5. Comparisons of The difference of intensity of acupuncture between the same group, in both wide type mice and TRPV1gene knockout mice, There was a significant difference each groups (P>0.05), while in CPZ mice, there was no significant difference among different intensity (P＞0.05).Conclusions:1. A stronger inhibition was showed in WT mice, suggesting the partial role of TRPV1involved in the modulation of gastric motility.2. EA at "CV12" exhibited an characteristic of intensity-dependence3.
