Abstract/Summary:
ObjectiveChronic hepatitis B (CHB) is a worldwide chronic liver disease. CHB is related toenvironmental fators,age, host fators, more thought to have close relationship with geneticfactors. Up to now,many domestic and foreign research points out that there is a closerelationship between the liver PNPLA3gene polymorphism and NAFLD, alcoholic liverdisease, hepatitis and other diseases.Howerer, there have yet been not reported research onthe link between PNPL A3gene rs738409polymorphism and chronic hepatitisB.Correlation at present. In view of the above reasons, in Qingdao Chinese Han populationin patients with chronic hepatitis B, we study the distribution of PNPLA3gene rs738409polymorphism, explore the association between PNPLA3gene polymorphism in QingdaoHan population andchronic hepatitis BMethodsThe study, thtough the colletion of the history and the arrange of biochemical dataresults,collected349cases, with185patients with CHB and164healthy control subjects.In CHB group and normal group, DNA was extracted, from the peripheral bloodpolymerase chain reaction (PCR) amplification of the target gene fragment sequences andgene sequencing methods to detect the PNPLA3genes rs738409polymorphism,thedistribution of genotypes and allele gene were determined by sequencing results.TheHardy-weinbeurg genetic equilibrium analysised the group representation of genotypes inCHB group and control group.non-conditional logistic regression analysised therelationship with genetic variation and CHB relevant factors. Data analysis of theapplication of statistical software SPSS17.0.0。Results Three genotypes are both sequenced in two groups: in CHB group GG:13.0%,GC:37.8%,CC:49.2%;in control group GG:6.7%,GC:30.5%,CC:62.8%The distribution ofgenotypes was significantly different in two groups. The frequency of rs738409G allelewas significantly different with31.9%in CHB patients and21.9%in control group (P<0.05).The study revealed an increased risk of developing CHB for rs738409G allelecompared with C allele (OR=1.67,95%CI:1.18-2.34, P=0.003). Logistic regression modelanalysis adjusted for confounding factors exhibited that carriers with the GG+GC genotypeof PNPLA3rs738409had increased risk of CHB compared with those with CC genotype(OR=1.76,95%CI:1.14-2.71, P=0.011)., to further show PNPLA3was associated with thedevelopment of CHB.ConclusionThe results revealed the rs738409G allele increases the risk of CHB in Chinese Hanpopulation in Qingdao,and its mechanism needs further study...
